The research outputs will include mechanistic investigations utilizing genome-wide CRISPR screens to identify and validate genes regulating bisphosphonate function and ONJ pathogenesis; characterization of relevant cell types and molecular drivers (including the lysosomal protein ATRAID) contributing to dysregulated cellular responses; and pharmacogenomic analyses of germline DNA from over 2,000 patients with metastatic bone disease enrolled in the NCI-funded clinical study SWOG S0702 to identify genetic markers and develop a polygenic risk score for ONJ susceptibility.
