Candidates will undergo minimal epitope definition, cross-reactivity screening, and in vivo benchmarking. In parallel, the recipient will enhance moderate-affinity receptors restricted by HLA-B07:02 and C07:02 using a function-first CDR3A mutagenesis platform that selects directly for T-cell activation under antigen-limiting conditions. The award addresses the clinical limitation that HLA restriction currently confines TCR-T eligibility largely to HLA-A*02:01, excluding many patients and enabling resistance via loss of the targeted HLA. The recipient, performing work in Newark, New Jersey, will deliver clinically actionable HPV16-specific TCRs suitable for TCR-engineered T cell therapy (TCR-T) and TCR-T cell engager therapy (TCR-TCE) applications. Performance extends through June 30, 2031.
