The National Institute of Allergy and Infectious Diseases (NIAID) awarded The University of Texas Southwestern Medical Center a Project Grant of $415,000 under the Allergy and Infectious Diseases Research program (CFDA 93.855) effective May 7, 2026, with a completion date of April 30, 2031. This investigator-initiated research project will generate scientific knowledge and methodological advances regarding the mechanistic function of a novel cell cycle regulator complex composed of two previously uncharacterized zinc finger transcription factors, ZFP574 and THAP12, and their role in regulating ribosome biogenesis and immune cell competence. The primary deliverables include: (1) mechanistic characterization of how the ZFP574-THAP12 complex supports immune cell development and function through regulation of POLR1H (a core RNA polymerase I subunit critical for ribosomal RNA synthesis); and (2) preclinical evaluation of the therapeutic potential of targeting this regulatory pathway in disease models of leukemia and autoimmunity. The research addresses a significant gap in understanding ribosome biogenesis regulation within the immune system, with direct public health relevance to preventing both malignancy and immune deficiency. Performance will be conducted at UT Southwestern's Dallas, Texas location.Federal Grant Award Summary
