The project centers on a novel mechanism called endovesiclosis, in which cationic-arginine-functionalized iron oxide nanoparticles bind therapeutic cargo electrostatically and enable passive internalization into extracellular vesicles without disrupting membrane integrity or compromising bioactivity. The embedded nanoparticles simultaneously enable non-invasive MRI-based tracking of engineered vesicles in vivo. The work will optimize nanoparticle formulations for efficient and safe loading across multiple cargo types, then evaluate therapeutic efficacy of miR-424- and CRISPR-SMURF1-loaded vesicles in a rat calvarial bone defect model. Therapeutic readouts include microcomputed tomography, histology, gene modulation, functional bone regeneration, biodistribution, and off-target analyses.
