The $1,200,000 award aims to enable confident identification of challenging classes of peptides for which current mass spectrometry (MS) methods fail or produce unreliable results. The project will develop new computational tools for two-dimensional mass spectrometry (2D-MS), which introduces fragment-fragment correlation (FFC) to dramatically improve the ability to reconstruct peptide sequences and pinpoint chemical modifications, even in complex or novel peptides. This work will target three long-standing challenges in proteomics: sequencing long, multi-charged peptides; de novo sequencing without reference sequences; and analysis of heavily modified peptides relevant to epigenetic regulation and drug development.
