BACKGROUND
The U.S. Department of Health and Human Services, National Institutes of Health, National Cancer Institute (NCI), Center for Cancer Research (CCR), Cancer Genomics Section, Genetics Branch intends to purchase a brand name or equal Epifluorescence Microscope and Imaging System for the purpose of obtaining very high-resolution data with extremely high sensitivity for the analysis of submicron extracellular vesicles (EVs) from Leica Microsystems Inc., Chicago, IL 60693.
The purpose of the instrument is to be able to perform studies that will identify and characterize single EVs and single fluorescent molecules across multiple lasers. This ultra-high level of sensitivity and precision is required for tests that will detect and enumerate EVs that have as few as a single molecule of interest. The purpose of this instrument is to provide single-molecule resolution for EV analyses as part of the EV analysis pipeline and the characterization of nanomaterials that are integral to the research that is being conducted in the Translational Nanobiology Section of the Laboratory of Pathology, as part of the wider NCI mission to generate and characterize new tools for cancer diagnosis and treatment. This instrument's specifications are particularly suited as an orthogonal means of cross-validating data from high-sensitivity flow cytometry platforms.
The purpose of this requirement is to procure and install a 333that will be used as part of the Epifluorescence Microscope and Imaging System suite of equipment for the Molecular Genetics Branch, CCR. A considerable proportion of our research program depends on the visualization of cancer relevant genes using fluorescence in situ hybridization (FISH). In addition, numerous colleagues in the intramural research program seek our expertise in determining chromosomal translocations and chromosomal copy number changes in human malignancies and murine models, and the localization of genes and chromosomes in the interphase nucleus.
