About this file

This Statement of Need (SON) describes the Epigenomics Core Facility's requirement to purchase a NextSeq 2000 Sequencing System for the National Institutes of Health National Institute of Environmental Health Sciences (NIEHS). The procurement is a Firm Fixed-Price Purchase Order to replace an aging NextSeq 500 sequencer, with specific technical requirements including maximum output of 540 Gb per run, 1.8B single reads, and 2 x 300bp read length. The system must include an embedded touchscreen monitor, on-instrument computer, control software, Dragen Bio-IT secondary analysis pipelines, installation, training, and a 12-month warranty.

Delivery is specified for Guang Hu at 111 TW Alexander Dr in Research Triangle Park, Durham, North Carolina, with items to be delivered within 10 weeks of purchase order award. The facility currently processes approximately half of its sequencing projects with NextSeq technologies and anticipates increased usage, necessitating the replacement of the older sequencer with a more advanced NextSeq 2000 system to meet the sequencing demand of NIEHS investigators.

STATEMENT OF NEED (SON)

1.0 TITLE

NextSeq 2000 Sequencing System

2.0 BACKGROUND

The Epigenomics Core Facility requests the purchase of a NextSeq 2000 sequencer to replace an old

NextSeq 500 sequencer. We currently have two NextSeq sequencers in the core: one NextSeq 500

(purchased in 2014) and one NextSeq 2000 (purchased in 2022). The NextSeq 500 is over 10 years old and is reaching its expected lifespan. It also provides lower throughput but higher operating costs than the NextSeq 2000 due to older technologies. The NextSeq sequencers process ~ 1/2 of our sequencing projects at the moment and the usage is expected to increase in the future. Therefore, a new NextSeq 2000 is needed to keep up with the sequencing demand by NIEHS investigators.

3.0 TYPE OF ORDER

This is a:

• Firm Fixed-Price Purchase Order

• Severable

4.0 PRODUCT FEATURES/SALIENT CHARACTERISTICS

Fragmentation-based molecular annotations are the gold standard in high-confidence biomarker identification using mass spectrometry. The new imaging parallel reaction monitoring parallel accumulation serial fragmentation (iprm-PASEF) workflow integrates CCS-enabled MALDI Imaging of lipids with a targeted MS/MS imaging approach that allows spatial fragmentation and analysis of multiple targets in a single acquisition. The workflow includes a fully integrated pipeline within our software ecosystem to generate accurate identifications with spatial fidelity.

The following product features/characteristics are required for this requirement:

• Automated generation of DNA clonal clusters by bridge amplification, sequencing, primary analysis and secondary analysis

• Maximum output per run 540 Gb

• Maximum single read per run 1.8B

• Maximum read length 2 x 300bp

• Embedded touchscreen monitor and on-instrument computer, control software, hardware accelerated Dragen Bio-IT secondary analysis pipelines

• Installation

• Training

• 12 month warranty
