Statement of Need Spectrofluorometer
Purpose: Acquisition of a new Agilent Cary Eclipse G9801AA spectrofluorometer or equal for spectroscopy analysis at the National Oceanic and Atmospheric Administration (NOAA), National Centers for Coastal Ocean Science (NCCOS) Beaufort NC Laboratory.
Background: The NCCOS Beaufort Laboratory requires one (1) new Agilent Cary Eclipse G9801AA spectrofluorometer or equal to be used for molecular spectroscopy to support satellite validation for ongoing HAB forecasting projects.
Specifications/Salient Characteristics: The Contractor shall provide one (1) Agilent Cary Eclipse G9801AA spectrofluorometer or equal which shall meet the following minimum specifications/salient characteristics:
• The instrument and accessories must include a full 1-year warranty.
• System installation and training by a certified service engineer must be included with the instrument.
• Computer-controlled, ratioing fluorescence spectrophotometer with dual monochromators and measurement modes for fluorescence, phosphorescence, chemiluminescence, and bioluminescence shall be switchable by software selection.
• The light source must be a pulsed Xenon lamp with a guaranteed life of at least two years.
The lamp must be on only during sample measurement to eliminate photobleaching effects and include a fully enclosed lamp module to eliminate light leakage.
• The light source shall include a narrow 2ms lamp pulse for peak power and sensitivity.
• The light source must be rated to last nearly the lifetime of the instrument.
• All reflective optics must be quartz over-coated to provide better sensitivity and use Schwarzchild collection optics.
• Operating range through Ultraviolet and Visible wavelengths shall be at least 200-900 nm (nanometers)
• In fluorescence mode, the instrument shall be able to complete full, high-speed scans across both excitation and emission spectra with a scan speed of 0.010-24,000 nm/min
• Rapid data collection shall be at least 80 Hz capable of data point measurement in <20 ms (milliseconds)
• Shielded to provide immunity from room lighting effects.
• Capacity to collect time resolved measurements.
• Excitation and Emission monochromators o 0.125 m Czerny-Turner design with an f3.6 focal length and have a limiting resolution of <1.5 nm.
o Monochromators must have fixed selectable slits with spectral band passes of 1.5, 2.5, 5, 10, and 20 nm plus 10 nm round and include as standard, computer-controlled stray light/scatter rejection filters.
o Monochromator Wavelength Range: 200-900 nm with an enhanced red-sensitive PMT detector, zero order selectable.
o Monochromator Wavelength Accuracy: ± 0.5 nm at 541.6 nm, ±1.5 nm over entire range.
o Monochromator Wavelength Reproducibility: ±0.2 nm o Maximum Slew Speed and Scan Rate: 24000 nm/min (400 nm/s) o Gratings: 30 x 35 mm, 1200 l/mm, blaze at 370 nm excitation and 440 nm emission.
• The instrument must include internal user-selectable filters on both the emission and excitation beams to exclude secondary light. As follows:
o Excitation includes open and shutter settings, 2 bandpass filters (250-395 nm and 335-620 nm), and 2 cutoff filters (550 nm, 695 nm).
o Emission includes open and shutter settings, 1 bandpass filter (250-395), 4 cutoff filters (295 nm, 360 nm, 430 nm, and 550 nm), and 3% attenuator.
• With the horizontal beam and slit image, the amount of sample required for analysis is as low as 500 µL(microliters) in a standard 3 mL cell.
• Sensitivity using the Raman band of water shall be at least 750:1 RMS using 350 nm excitation, 10 nm slits, 1 s signal averaging; 500:1 RMS using 500 nm excitation, 10 nm slits, 1 s signal averaging.
• Sample and reference signals shall be detected by high performance photomultiplier tubes (PMTs) with enhanced red-sensitive R3896 PMT to provide significant signal improvement in the 600 to 900 nm range.
• The PMTs shall be able to operate with a gain of Low, Medium, or High, with manual adjustment from 400-1000 V in increments of 1 V.
o Data shall be collected using a phase stepping wavelength drive to eliminate scan speed errors by synchronizing the lamp pulse and scan motors so that no measurements are taken while the monochromators are moving. The time drive mode also synchronizes the lamp pulse to data acquisition.
• Allowable data intervals must include: 0.15-30 nm. Data collection rates in points per minute per cell are 4800 points/1 cell; 6 points/4 cells; and 55 points/4 cells with minimum SAT and Dwell Time.
o Signal averaging varies with the selected collect mode as follows: Fluorescence (0.0125 to 999 s), Phosphorescence (1 ms to 10 s Gate time), Bio-Chemi- Luminescence (40 ms to 10 s Gate time)
• Includes Single cell holder (1 cm rectangular cells)
• Instrument <25L x 25.5D x 12H inches wide due to limited space availability
• Power: 100-240V AC
• Shall include control and analysis software compatible with Windows 10 operating systems in keeping with current NOAA IT requirements but shall NOT include a computer (NOAA IT will provide).
• Includes Microplate Reader Accessory:
o Reader shall provide full 10-wavelength scanning using reflective optics instead of fiber optics.
o Shall include reader, standard, 10mm x 10mm holder, sample compartment door o Shall include installation and familiarization of plate reader accessory.
o Shall install with a lock-down mechanism on the base plate that does not require any tools.
o High speed data collection from 96-well plates in < 1 minute.
o Software shall allow single wavelength measurements, multiple wavelengths, wavelength scanning and continuous kinetic modes for plates.
o Reader shall have an adjustable image spot down to 2mm in diameter.
o Reader shall auto-align beam on microplate in ≤1 minute.
Instrument must be capable of being fitted with the following optional accessories for future capacity:
o Automated Polarizer o Fiber Optic cable for additional data collection flexibility o Solid Sample Holder o Probe for precise temperature monitoring o Thermostatted Cell holder and Circulating Water Bath
Software: The instrument shall include control software that meets the following requirements:
• The instrument must be PC controlled, and the software must be supported by Microsoft Windows 10 operating system.
• Applications must include at least Scanning, Thermal, Ratio, Simple and Advanced Reads, Concentration, Kinetics, and Lifetimes.
• Must provide a built-in applications development programming language to allow customization to fit unique or special requirements.
• A “Help” function must provide easy access to the entire operation manual; clicking on the help for a specific screen should take the user to help for that particular screen.
• User must be able to access and manipulate the fluorescence data using the instrument software, without being online with the instrument.
IT Security Requirements
• The Assessment and Authorization (A&A) requirements of Clause 48 CFR 1352.239-72 do not apply, and a Security Accreditation Package is not required.
• The contractor will not access, store, process or transmit any government-privileged information. The contractor will not have access to any non-public government information, systems and/or networks.
Delivery Schedule: All components shall be delivered to the Beaufort Laboratory no later than 30 September, 2023.
