STATEMENT OF WORK

C. GENERAL INFORMATION

A. Introduction The United States Geological Survey (USGS) Upper Midwest Environmental Sciences Center (UMESC) is seeking services to conduct a Good Laboratory Practice (GLP) OPPTS 830.1700 Preliminary Analysis study of five samples of Antimycin-A (ANTA) material (inclusive of identification and quantitation of all compounds present at or above 0.1% w/w ) and five samples of ANTA integrated system (formulated product). Parameters will include precision and accuracy.

The prospective analytical analyses are necessary to support product registration with the U.S.

Environmental Protection Agency.

B. Background

ANTA is a fermentation product of Streptomyces sp. that has been used extensively as a pesticide for fish removals and fisheries management. The pesticide is a non-selective toxicant that affects cellular respiration and produces a lethal effect in fish at parts per billion concentrations. It is an ideal candidate for chemical fish removal applications because it does not impart a detectable taste or odor (to fish) and is rapidly degraded via hydrolysis and sunlight leaving no known toxic residues.

The quantitation of total ANTA is performed on a HPLC-MS system. Antimycin-A samples contain a mixture of multiple stereoisomers. The four most abundant pairs of stereoisomers, specifically A1- A4, are quantified per sample. Going forward, the sum of these four pairs, ANTA(A1-A4) will be referred to as total ANTA, or ANTA.. ANTA is the active ingredient in a pesticide that the USGS is currently pursuing to register with the U.S. Environmental Protection Agency (USEPA).

C. Definition/Applicable Documents

Figure 1. The active ingredient, Antimycin-A, general chemical structure and the variable groups are indicated by R (e.g., R1, R2). These variable groups are the subcomponents that vary between stereoisomers.

PAGE OF PAGES

D. Scope The independent laboratory will prepare and conduct a GLP compliant OPPTS 830.1700 Preliminary Analysis study (also known as a 5-batch study) of ANTA material (containing purity and impurity quantification), and a OPPTS 830.1700 Preliminary Analysis study of the fully formulated ANTA pesticide (purity quantitation only) and prepare a report for each of the study findings.

D. WORK REQUIREMENTS

A. Technical Requirements

Below is a list of specific reagent(s) and apparatuses necessary for the ANTA analytical method.

Reagents:

• Acetonitrile, HPLC grade or better

• Ethanol, HPLC grade or better

• Water ≥18.0 megaohms-cm or similar equivalent

• Methanol, LC-MS grade

• Isopropanol, LC-MS grade

• Ammonium acetate, ACS grade

• Anhydrous Sodium sulfate, anhydrous, certified A.C.S.

• Acetone, HPLC grade or better

• Formic Acid, ACS Reagent grade or better

Apparatus: An LC-MS system described below:

• Agilent 1290 Quaternary Pump (G7104A), 1290 Multi Column Temperature control

(G7116B), 1290 Multisampler (G7167B), 1290 Variable wavelength detector (G1314E), QQQ LC/MS (G6460A) and Agilent Mass Hunter software for system control or equivalent LC-MS system.

• LC column, Phenomenex Kinetex 1.7µm particle size, XB-C18 100Å, 100 × 2.10

Laboratory equipped with the following, but not limited to:

• LC vial caps and septa, matching cap with Teflon®/silicon septa; Waters LC-MS vial caps or equivalent product

• Minus 20 degree Celsius freezer for analytical standard and sample storage

• Powered Air Purifying Respirator (PAPR)

• Balance capable of measuring to ±0.02 mg

The independent laboratory will prepare protocols and conduct two GLP 5-batch studies, one of ANTA material (purity and impurity) and one for ANTA final formulated product purity.

Parameters will include precision, accuracy, standard curve linearity, and specificity. Four batches of each ANTA material and ANTA formulated product will be prepared in duplicate and one batch each will be prepared 5 times for purity and precision determination. Impurity studies on 5-batch ANTA material must include the determination of all impurities present at 0.1% or higher using a low ANTA concentration (e.g., 3-5 µg/L) per batch.

PAGE OF PAGES

The analytical instrument used for the 5-batch ANTA material purity and ANTA integrated system purity determination must be equivalent in sensitivity (e.g., response) to a High Performance Liquid Chromatography coupled with Triple Quadrupole (QQQ) mass spectroscopy (HPLC-QQQ). The HPLC-QQQ operating conditions for acquisition of ANT-A(A1-A4) will be provided to the independent laboratory upon award of the service contract. The ANTA(A1-A4) calibration curve should consist of at least five points ranging from 1 - 400 µg/L.

The final report for ANTA material must include the 5-batch ANTA material purity and impurity determination, including the number of impurities present at 0.1% or higher. The final report for the ANTA formulated product must include purity determination of the 5-batch ANTA formulated product. The reports must include the ANTA standard calibration curve performance characteristics including results compared against the following acceptance criteria: standard calibration curve (total ANT-A(A1-A4) R2 ≥ 0.995 linearity), retention time (k’ between 3-8, variability ± 5%), QC standard check within ± 20% of theoretical concentration, instrument precision (percent relative standard deviation (%RSD < 20%) and accuracy (each standard ± 20% theoretical concentration), technician precision (n=3 samples %RSD < 20%), percent and peak symmetry results ≥ 0.55 for all peaks but less than 2.0.

1. Criteria for acceptance of Product or Service

Two completed final reports in compliance with GLP and the approved protocols with no deviations considered critically detrimental to the study findings.

B. Deliverables

1. Characteristics for work product described in II. A. and A. 1. Identified by name and quantity.

Final reports with details as listed above in compliance with GLP.

2. Delivery dates for work/product to be completed and point of delivery.

08/1/2023 – 08/1/2024, dependence on ANTA material manufacturing; however, following 5-batch ANTA material and formulated product purity and impurity studies, the final report must be received within 30 days of analytical acquisition completion.
