I. GENERAL INFORMATION

A. Introduction

The United States Geological Survey (USGS) is requesting bids for 96-well enzyme-linked immunosorbent assay (ELISA) kits for the analysis of the cyanotoxins anatoxin-a, cylindrospermopsin, microcystins/nodularins, and saxitoxin.

B. Background

The USGS New York Water Science Center (NYWSC) has developed the capability for cyanotoxin analysis using a Cyanotoxin Analysis Automated System (CAAS). The CAAS automates laboratory analysis steps for cyanotoxin analysis by enzyme-linked immunosorbent assay (ELISA) using 96-well plates. Ongoing studies in the NYWSC require ELISA analysis of the following cyanotoxins: anatoxin-a, cylindrospermopsin, microcystins/nodularins (ADDA, using EPA Method 546), and saxitoxins. These analyses require assay kits that contain 96-well plates coated with antibodies relevant to each cyanotoxin, as well as associated standards and reagents. Overnight shipping in chilled coolers is required to ensure integrity of the kits and standards. Without the required kits and reference standards the NYWSC will be unable to perform the analyses required to meet project needs.

C. Definitions/Applicable documents

U.S. Geological Survey National Field Manual for the Collection of Water-Quality Data Chapters: A7.4 Algal

Biomass Indicators and A7.5 Cyanobacteria in lakes and reservoirs: toxin and taste-and-odor sampling guidelines (https://water.usgs.gov/owq/FieldManual/Chapter7/index.html).

D. Scope

The USGS requests bids for 96-well ELISA kits for the analysis of the cyanotoxins anatoxin-a, cylindrospermopsin, microcystins/nodularins, and saxitoxin. This will be a 5 year contract. A minimum of 5 kits per cyanotoxin ordered per year. A maximum of 50 kits for each cyanotoxin kit ordered per year. Overnight shipping in chilled coolers is required.

II. WORK REQUIREMENTS

A. Technical Requirements:

It is the intent of these specifications to describe the general requirements for analysis of the cyanotoxins anatoxin-a, cylindrospermopsin, microcystins/nodularins (ADDA, using EPA Method 546), and saxitoxins by

ELISA.

1. Anatoxin-a Analysis (all items on this list must be included as specified).

• Monoclonal direct competitive 96-well plate ELISA assay suitable for quantitative detection in water samples

• ELISA assay kit will include: coated 96-well microtiter plate, lyophilized anatoxin-a-HRP enzyme conjugate, conjugate diluent, lyophilized anti-anatoxin-a antibody, antibody diluent, empty clear and amber HDPE bottles, anatoxin-a standards (0, 0.15, 0.4, 1.0, 2.5, and 5.0 μg/L), control sample (0.75 μg/L), sample diluent, wash buffer, substrate solution, and stop solution

• Test detection limit of 0.1 μg/L

2. Cylindrospermopsin Analysis (all items on this list must be included as specified).

• Direct competitive 96-well plate ELISA assay suitable for quantitative detection in water samples

• ELISA assay kit will include: coated 96-well microtiter plate, cylindrospermopsin standards (0, 0.05, 0.10, 0.25, 0.50, 1.0, and 2.0 μg/L), control sample (0.75 μg/L), sample diluent, cylindrospermopsin-HRP conjugate solution, antibody solution, wash buffer, substrate solution, stop solution

• Test detection limit of 0.04 μg/L

3. Microcystin Analysis (ADDA, Using EPA Method 546) (all items on this list must be included as specified).

• Indirect competitive 96-well plate ELISA assay suitable for quantitative congener independent detection in water samples

• The assay must be certified by the USEPA Environmental Technology Verification Program for use with EPA

Method 546

• ELISA assay kit will include: coated 96-well microtiter plate, microcystin standards (0, 0.15, 0.40, 1.0, 2.0, and 5.0 μg/L), control sample (0.75 μg/L), sample diluent, antibody solution, anti-sheep-HRP conjugate solution, wash buffer, substrate solution, stop solution

• Test detection limit of 0.10 μg/L

• Coefficients of variation for standards of <10% and for samples <15%

• Assay must exhibit good cross-reactivity will all cyanobacterial cyclic peptide toxin congeners tested to date

4. Saxitoxin Analysis (EC 2002/225 Compliant) (all items on this list must be included as specified)

• Direct competitive 96-well plate ELISA assay suitable for quantitative detection in water samples

• ELISA assay kit will include: coated 96-well microtiter plate, saxitoxin standards (0, 0.02, 0.05, 0.1, 0.2, and

(c) If the Government fails to place orders for the minimums computed under paragraph (a), above, the contractor will not be entitled to payment of the contract price for the unordered quantities, but will be entitled to damages suffered as a result of the Government's failure to order the minimum quantity.
