GENERAL INFORMATION

1. Scope

The USGS Patuxent Wildlife Research Center is requesting global (untargeted) metabolomics analysis of ceca and plasma samples, analysis and interpretation of resulting data, including annotation of the data with relevant metabolic pathways. Global metabolomics analysis is a method for analyzing metabolic products, which does not target specific metabolites, but rather provides a broad-based screen of thousands of chemically and structurally diverse metabolites. The metabolites measured shall include metabolites from each of the following classes: amino acid metabolism, cofactor and vitamin metabolism, nucleotide metabolism, microbiome metabolism, carbohydrate metabolism, energy metabolism, lipid metabolism, and other novel metabolites.

WORK REQUIREMENTS

1. Technical Requirements

• Vendor must have access to a reference library of metabolite signatures containing more than 12,000 metabolite profiles, the majority (>2/3) of which are not publicly available but have been authenticated in-house.

• The vendor must have a demonstrated record of project completion, including those for federal government research laboratories, in multiple sample matrices including plasma, and contribution to peer-reviewed journal publications.

• The vendor must employ an automated data handling, analysis and quality assurance process for systematic sample management, which includes o 1) LIMS designed specifically for mass spectrometry-based metabolite analysis o 2) Automated mass spec data extraction and reduction software with chromatographic peak alignment tools for compound identification o 3) A comprehensive spectral library for the identification of analytical signatures

• The vendor must have at least 5 to 10 years of experience in whole-metabolome profiling

• The required volume for samples must be 100 microliters (µl) or less per sample

• The vendor must store samples immediately in -80 degrees until ready for assays

• Analyses will include the use of ultra-high performance liquid chromatography/tandem accurate mass spectrometry (UHPLC/MS/MS)

• The median spectral relative standard deviation, indicative of analytical reproducibility (instrument variability) must be 55%

2. Deliverables

• Raw data, a comprehensive report that includes detailed analytical methods, summarized QC results, relevant statistical analysis of data, representative figures (e.g. heatmaps), biological interpretation and literature support

• Must include Quality Control (QC) samples in the analyses

140G0120Q0140 Page 5 of 27

• The following QC samples must be analyzed intermittently throughout the analytical experiment (a minimum of 20% of injections/day) and results reported to USGS:

o extraction/solvent blanks (extraction negative control) o process blanks (instrument/analytical negative control) o pooled matrix (study) samples (unknowns) o pooled reference samples (previously characterized performance evaluation samples) o unknown sample spikes (internal standards= pre-extraction spikes) o recovery standards (post-extraction, pre injection spikes)

• Bioinformatic analysis of raw data and subsequent generation of relative concentration profiles must be provided.

• Statistical analysis of generated data must be provided to USGS

• Residual samples, if any, will be stored in the laboratory at -80 degrees Celsius. All

• Biological materials provided by the USGS, all residual specimens, and all data generated shall be the property of the USGS and shall be returned to the USGS after the assays are completed and data approved.

• Turnaround time of 14 weeks or fewer from the date of sample submission

• The Contractor shall notify the principal investigator within 72 hours of any incoming shipments received in a damaged condition

3. Quantity Maximum

Orders will not exceed 750 samples over a 5-year period.
