Statement of Work (SOW) Project: Dairy Cattle Genomics and Transcriptomics
GENERAL INFORMATION
1.0 Scope of Work: Several research programs here at Dairy Forage Research Center are presently or evolving to perform more sophisticated next generation sequencing (NGS) projects. Due to the objectives of our CRISs, our research projects include cattle genomics and transcriptomics study using both short reads, targeted amplicon capture and long reads sequencing. Additionally, in terms of targeted tissue types and research organisms, our research programs currently cover a wide range of tissues types and organisms and are rapidly expanding. These include cattle tissue specific RNA sequencing, rumen microbial community sequencing and comparative genomics, forage crop transcriptomics and genomics analysis at population level, and the interaction between host and rumen microbial community using single cell sequencing. To fulfill the research needs, we purchased the PerkinElmer Sciclone to help with various next-generation sequencing library preparations.
2.0 Background: after the factory warranty expired, we have purchased one full-year of service contract covering April 20, 2019-April 19, 2020. In order to ensure the smooth operation of the instrument, we foresee future continuation of service contract.
CONTRACTOR REQUIREMENTS
3.0 Technical Requirements/Tasks: The service contract will perform repair and calibration on both hardware and software on the Sciclone. The detailed description of the task is listed below.
o
1. Provide repair on the hardware to ensure the maintain the following function of the instrument
• 96 Channel Head for Aspirating and Dispensing of reagents and samples
• Volume range of the 96 channel head from 1-200ul
• Process single well, single column, single row or complete plate
• Three 4-100°C temp controlled locators
• One 4-70°C shaking locator
• Magnetic bead locator
• Microplate gripper for automated consumable management
• Four-sided environmental enclosure with access doors: prevent access and preserve sample integrity during a running assay. Opening the doors of the enclosure results in an immediate soft stop of the robot motions which may be resumed upon closing the doors.
• Instrument enclosure also supports either an optional UV light, HEPA filtration device and/or an active (fan) external fume port.
• Optional LV and NL aspirating & dispense heads
• Optional 8 channel (single well) pipetting option
• Optional barcode reader wand and/or deck mounted reader
• 24 position deck with 4 axis motion control (XYZ/gripper)
• Plate and tip box disposal chute accessible by gripper
• On deck tip disposal chute
2. Provide repair on Gripper:
• To ensure the proper function of the gripper that is independent of the multichannel pipetting head, and that enables the instrument to pick up a microplate lid while simultaneously carrying out pipetting operations with the multi-channel pipetting head.
• To ensure The gripper has the ability to stack, de-stack, and transfer plates/equipment among all 24 deck positions.
• To ensure the proper function of a built-in detector which confirms when a plate has been successfully gripped, and if one has not been gripped or has become dislodged, the software pause the instrument and initiate an error condition.
3. Provide technical support on Sciclone G3 NGS Workstation Software
• To ensure that Maestro software operates the Sciclone G3 NGS Workstation and coordinates all operational steps
• To ensure the Application Assistant features operates with an intuitive user interface and simplified protocol setup to support users of all levels
• To ensure the proper function of E-mail notification of status
• To ensure the continuation of unattended runs when connected to the internet
4. Ensure the LIMS and Analysis Compatibility
• To ensure and maintain the integrated barcode reading capabilities to barcodes on sample, reagent, and other receptacles
• To ensure the ability to write log files with sample tracking information to network folders, specified directories to be uploaded and parsed to database tables o
The contractor performs the repair and calibration on the Scicone without government supervision. Specify requirements clearly so all readers can understand the content. The service will ensure the instrument operates at the original, factor level performance.
4.0 Government Furnished: Government will reviewing the repair work completed at the end of service repair visit and sign off the report. The facility is located at 1925 Linden Dr.
Madison, WI. It is open Monday-Friday, from 8-4PM.
