SB1341-15-RQ-0953

Statement of Work

I. BACKGROUND INFORMATION

The National Institute of Standards and Technology (NIST), Cell Systems Science Group, requires a quantitative phase imaging camera that can be attached to an existing optical microscope and perform live-cell imaging.

NIST needs to track single living cells and monitor dry mass evolution over time for these single cells. This capability is required for several cell types including bacteria and stem cells, with the result being repeatable, quantitative mass. This imaging capability is being developed for applications that are critical programmatic areas for the division.

For both applications and theoretical understanding, robust and repeatable quantitative phase images must be achievable across multiple optical microscopes, both inverted and upright. An interference method of quantitative phase, such as quadri-wave lateral shearing interferometry, in a camera that adapts to any microscope with minimal set-up time needed can be used to achieve this.

II. SCOPE OF WORK

This procurement request is for a quantitative phase imaging camera that must interface with the Cell System Group’s existing Zeiss Axiovert 200M optical, Zeiss Axioplan 2 optical, Olympus IX-70 and Olympus IX-71 optical microscopes. The Contractor shall also provide installation and training as described herein.

III. SPECIFICATIONS

The Contractor shall provide one (1) quantitative phase imaging camera and operating software meeting the following minimum technical specifications.

Only new quantitative phase imaging cameras shall be considered.

The quantitative phase imaging camera shall be able to attach to all of the following microscopes via a standard C-mount coupling:

o Be able to attach to a side port of a Zeiss Axiovert 200M, Olympus

IX-70, and Olympus IX-71.

o Be able to attach to the top port of the following upright microscope: Zeiss Axioplan 2.

The device shall acquire live phase images. The image intensity values shall consist of numerical values that are phase or optical thickness units and these units can be converted into real thickness values.

Quantitative phase images shall contain only positive values equal or greater than zero or negative values less than or equal to zero. The quantitative phase images shall be free of residual artifacts of generic phase contrast images that contain both positive and negative phase values.

Device shall have an acquisition rate greater than or equal to 1 Hz.

The device shall be compatible with any selected or filtered wavelength within the visible spectrum for the following illumination sources:

halogen, LED, and laser.

The device shall be compatible with white light from a halogen illumination source.

The device shall be compatible with both plastic and glass dishes used as live-cell sample containers.

The device shall have a length less than 30 cm / 11.8 inches.

The device shall have a width and height less than 15 cm / 5.9 inches.

The contractor shall provide software required to control the phase imaging camera.

o The software shall be Windows compatible (XP/7).

o The software shall be able to calculate the dry mass (pg) of specimens such as cells.

o The software shall be able to automatically track single cells over time for at least 24 hours.

o The software shall allow the camera to be operated from C++, Matlab and LabView.

o The software shall allow the camera to be driven with Micro-Manager

Open Source Microscopy Software.

The system shall include an operations and maintenance manual covering proper operation, routine maintenance, and troubleshooting for the instrument and controlling software. The manual shall be supplied in both hard copy and electronic PDF format to the NIST Technical Point of Contact.

Installation:

The Contractor shall install the camera in Building 227, Room A162, at NIST Gaithersburg, MD. Installation shall include, at a minimum, uncrating/unpacking of all equipment, set-up and hook-up of all equipment, start-up, demonstration of specifications, and removal of all trash.

Instrument shall operate to NIST’s specification upon completion of installation. Installation must take place during normal business hours, between 8:30 am and 5:00 pm Eastern Time, Monday through Friday except Federal Holidays, and must be coordinated with the NIST Technical Point of Contact (TPOC). Installation must be completed within 60 days of award.

Training:

The contractor shall provide training at NIST for up to 2 NIST personnel covering normal operation, troubleshooting, and routine maintenance.

Training shall be provided during normal business hours, between 8:30am and 5:00pm Eastern Time, Monday through Friday, except Federal Holidays, and shall be coordinated with the NIST Technical Point of Contact (TPOC) to ensure maximum availability of NIST personnel. Training shall be completed within 10 days of installation. The training may be completed on-site at NIST immediately after installation and demonstration of specifications.

Acceptance Testing:

Acceptance testing shall occur not later than five days after successful completion of delivery, installation and training. For the acceptance testing, trained NIST personnel will install the device on each microscope specified in this Statement of Work to ensure that the device mounts correctly and interfaces correctly with each microscope. Additionally, trained NIST personnel will install the device on the NIST owned Zeiss

Axiovert 200M microscope using the C-mount coupling and perform the following tests:

Validation of real thickness values. The NIST TPOC will measure the real thickness value of a NIST provided test sample of a microsphere with a known diameter and refractive index. The Government requires that the known diameter and the measured real diameter shall register a difference of less than 5%.

Validation of Live Cell Tracking. The NIST TPOC will set up a live cell time lapse acquisition in usual growth conditions on the Axiovert 200M microscope and test the device’s ability to track live cells for up to 24 hours.

Upon successful completion of testing, the Government will provide official acceptance of the system in writing via email to the contractor.

Any deficiencies discovered during the acceptance testing will be communicated to the contractor via email by the NIST TPOC. It shall be the contractor’s responsibility to remedy any deficiencies including repairing and/or replacement of the system at no additional cost to the Government.

Warranty:

The Contractor shall provide, at a minimum, a one year warranty for the equipment, both hardware and software. The warranty shall cover all parts, labor and travel. The warranty shall commence upon successful completion of delivery, installation, training and acceptance.

IV. PERIOD OF PERFORMANCE

Delivery, installation, training and acceptance testing shall be completed not later than 85 days after receipt of order.

The items shall be shipped F.O.B. Destination to: National Institute of Standards and Technology, Building 301 Shipping and Receiving, 100 Bureau Dr., Gaithersburg, MD 20899.

V. PLACE OF PERFORMANCE

All work shall be completed at the Contractor’s facility. Installation of the instrument and training shall be accomplished at NIST, Gaithersburg, Maryland. Normal duty hours are Monday through Friday, 8:30 a.m. to 5:00 p.m. with the exception of Federal holidays.
