Purchase Description

Human cytotrophoblast is an essential cellular component for building in-vitro three-dimensional vascularized placental tissue for studying infection of various external living microorganisms and viruses. NCATS DPI tissue bioprinting lab started the development of the placental barrier model with the seed grant of research-grade cells and tissue from the Amnion Foundation in 2020, and we have been using cells from the foundation. It has been challenging to secure cytotrophoblasts to improve robustness of the protocol for engineering 3D vascularized placental tissue to model virus infections such as SARS-COV2 and ZIKV. Thus, this acquisition of cytotrophoblsts, which we have confirmed compatibility with the our protocol development, allows us to improve our tissue quality control and expand our current 3D placental barrier tissue models for drug development.

NCATS Tissue bioprinting lab has developed a protocol for 3D vascular tissue model containing endothelial cells, pericytes, and fibroblasts. We develop the placental barrier tissue by addition of cytotrophoblasts on the vascularized tissue model. The inclusion of the cytotropholasts will allow for measuring barrier functions responding to various environmental cues. NCATS Tissue bioprinting lab has been using the cytotrophoblasts for a year and confirmed that it met the desired outcomes.

Human cytotrophoblasts

1) Brand name: Human cytotrophoblasts

2) Manufacturer’s name: Amnion Foundation

3) Model, type, catalog number: 1230

Cytotropoblasts were isolated from a full term placenta (after delivery of the baby).

Cytotrophoblasts (CTBPRO2®) available through the Amnion Foundation are immunopositive for CD49f and express both the epidermal growth factor receptor (EGFR) and cytokeratin 7 (CK7). They also secrete human chorionic gonadotropin (hCG) in culture.

Cytotrophoblasts are capable of differentiation into Syncytiotrophoblasts.

Cytotrophoblasts are grown in high oxygen tension.
