NIST REQUIRMENTS
TITLE: Automated fluorescence microscope LAB REQUESTING SERVICE: Physical Measurement Lab
I. BACKGROUND AND OBJECTIVES
The Biophysical and Biomedical Measurement Group within the Microsystems and Nanotechnologies Division of the Physical Measurement Laboratory, in coordination with the NIST-on-a-chip program, conducts research on miniaturized, “lab on a chip” (LOC) technologies with embedded sensors. The Group is developing microfluidic devices to measure dynamic responses of living mammalian cells and requires tools to optically interrogate cells and fluids. To accomplish this work, the Group seeks a fully automated fluorescence microscope that can be used to conduct fluorescent bioassays both in multiwell plates and custom microfluidic devices. Experiments will include counting and intensity-based measurement of fluorescent objects, time-lapse fluorescence imaging of fluorogenic reactions, cellular metabolism, and fluorescent protein production/degradation.
II. SCOPE OF WORK
The Contractor shall provide an automated microscope complete with phase and fluorescence imaging. Because the system must be compatible with existing Carl Zeiss microscopes and accessories in the lab, the vendor must provide Carl Zeiss system components or demonstrate compatibility with Carl Zeiss software control and component recognition.
III. Minimum Requirements:
All items must be new. Used or remanufactured equipment will not be considered for award. Experimental, or prototype will not be considered. The use of “gray market” components not authorized for sale in the U.S. by the proposer is not acceptable. All line items shall be shipped in the original manufacturer’s packaging and include all original documentation and software, when applicable.
Contract Line Item Number (CLIN) 0001: The Contractor shall provide one (1) Carl Zeiss automated microscope complete with phase and fluorescence imaging in accordance with the following minimum requirements:
· Zeiss Axio Observer 7 body with transmitted light and phase condenser, and fully motorized components, including focus drive, optics, and filters
· Requirements:
· 2.5x 0.085 NA
· 5x 0.16 NA (phase)
· 10x 0.3 NA (phase)
· 20x 0.35 NA (phase)
· Filter sets for filter wheel to include common fluorophore spectra (e.g. DAPI, eCFP, FITC/GFP/Fluo4, TRITC/Cy3, Alexa568, Cy5, and Cy7) and 4-color multi-pass filter set for simultaneous visualization of DAPI+GFP+TRITC+Cy5
· High resolution imaging system with 2 cameras (equivalent to Axiocam 712 mono and Hamamatsu ORCA Fusion)
· Black and white camera (12 Mpix) with 14-bit, 20 fps high-resolution acquisition
· Black and white camera (4 Mpix) with 16-bit, 89 fps full frame acquisition, selectable for continuous streaming of >1000 fps when limiting readout size, quantum efficiency > 70%
· We do not require software, a fluorescence light source, a motorized XY stage, or a incubation system.
IV. Delivery
Delivery shall be FOB destination and shall occur not later than three (3) months ARO
FOB Destination means: The contractor shall pack and mark the shipment in conformance with carrier requirements, deliver the shipment in good order and condition to the point of delivery specified in the purchase order, be responsible for any loss of and/or damage to the goods occurring before receipt and acceptance of the shipment by the consignee at the delivery point specified in the purchase order; and pay all charges to the specified point of delivery. The contractor shall deliver all Line Items to:
The National Institute of Standards and Technology 100 Bureau Drive, Building 221 Room A120 Gaithersburg, MD 20899-1640 V. Installation The contractor shall install the microscope at NIST location (listed above, subject to change). The contractor may be present during testing by NIST staff to assess that the products meet the contract specifications. Contractor must arrange this activity no later than 30 days after delivery and at their own expense. All onsite installation and demonstration shall be done at NIST, Gaithersburg, MD.
VI. Training
One training session for up to 6 associates at NIST Gaithersburg following installation during normal business hours within 30 days of delivery.
VII. Inspection and Acceptance In addition to the inspection and acceptance terms articulated in 52.212-4, the Government reserves the right to perform performance tests and evaluations to verify specified system performance. Such tests and evaluations, if performed, shall be conducted within the environment that the system is to be operated. The Contractor has the right to be present during the tests and evaluations of the technical specifications, at the Contractor’s expense.
Inspection and acceptance shall occur within ten (10) business days after delivery and installation of the complete system at NIST.
A visual inspection of the system/part (s) will be performed by the NIST COR to identify surface defects or any form of indication that the system/part (s) were damaged during transport to NIST. The Government shall have sole discretion to require repair or replacement of damaged and/or nonconforming supplies at no cost to the Government. The Government at any time prior to acceptance shall reject the system/part (s) due to defects and/or nonconformance.
Acceptance will be based on validation of the following:
· specifications of the imaging system, optics, and fluorescence components
· software control of system components
· Successful setup and acquisition of a multi-region, multi-channel timelapse imaging experiment
VIII. Place of Performance All work shall be accomplished at NIST, Gaithersburg, Maryland. Normal duty hours are Monday through Friday, 8:30 a.m. to 5:00 p.m. with the exception of Federal holidays.
IX. Government Furnished Property The government will provide Windows-based computer system and laser table for mounting the microscope.
NIST will provide a high-performance computer with the following specifications (or equivalent):
| Dell Precision 7920 Precision Workstation |
| Microsoft Windows 10 x64-bit Enterprise |
Intel Xeon Gold 6230 2.1GHz,(3 .9GHz Turbo, 20C, 10.4GT/s 3UP I, 27.5MB Cache, HT (125W) DDR 4-2933 1st) 8 GB NVIDEA Quadro video card 1 TB Solid Stage Drives (SSD) for operating system 8 TB SATA drive storage (minimum) 64 GB RAM (minimum)
X. Warranty The contractor shall warrant the entire system for a period of a minimum of 1 year after receipt of the equipment and shall be in accordance with terms in FAR 52.212-4
1. The Contractor shall provide a one (1) year warranty on all labor and parts for the microscope
2. Scheduling of the warranty service visits shall be consistent with the maintenance requirements of the instrument as detailed in the service and use manuals for the equipment.
3. Warranty service visits shall occur during NIST’s regular business hours as specified in Section XII of this SOW. The Contracting Officer’s Representative (COR) or the NIST Task Manager shall contact the Contractor to schedule a mutually acceptable date and time for the warranty service visits. The scheduled maintenance visits may be made at the same time as an unscheduled visit for corrective or other action if such a visit occurs within one month before a scheduled warranty service visit.
XI. Payment Schedule The Contractor shall be paid, in accordance with Net 30-day payment terms, upon receipt and acceptance of a proper invoice, in accordance with the following schedule:
