STATEMENT OF WORK
Protein Micro Array Testing at Antigen Discovery Inc.
1. Purpose & Objectives of the Procurement:
a. Description of the project: The Laboratory of Malaria Immunology and Vaccinology (LMIV) proposes for an LMIV scientist to be trained in protein micro array analysis at the vendor location over a period of four weeks. The hands-on training will include testing of LMIV test articles. The training will provide the scientist expert knowledge and proficiency such that future protein micro array testing will be conducted at LMIV facilities, and other scientists will be trained by our new expert.
b. We propose to bring the protein microarray technology to our lab and to the malaria community. Protein microarray is a useful technology that helps to screen humoral responses to various antigens simultaneously.
c. We plan to use a protein microarray chip containing 823 reactive proteins, selected after extensive serological evaluation of a larger proteome array, to seroprofile serum samples from about 200 pregnant mothers at different parity of placental malaria infection. The 823 proteins on these microarrays were expressed by in vitro trancription/translation using an E.coli cell free expression system and printed onto nitrocellulose pads. The serum samples will first be screened based on the parity of mothers, malaria severity and their delivery dates. Then the antibody reactivity to these 823 known and hypothetical Pf proteins will be assessed.
d. This approach will provide a unique platform to rapidly profile antibodies to malaria and suggest novel insights into the kinetics of immunity to placental malaria and identify Abs against known and hypothetical Pf proteins that may be associated with placental malaria. Furthermore, this strategy when validated in larger studies, including large epidemiological settings, may reveal proteins that could be targeted as potential placental malaria vaccines.
