Response Date: 06/20/2016

Classification Code: 68 – Chemicals & Chemical Products

NAICS Code: 325414

Contracting Office Address

Department of Health and Human Services, National Institutes of Health, National Cancer

Institute, Office of Acquisitions, 9609 Medical Center Drive, Room 1E128, Bethesda, MD

20892, UNITED STATES.

Description

The National Cancer Institute (NCI), Vaccine Branch, plans to procure on a sole source basis clinical grade TARP (TCR Alternative Reading Frame Protein) peptides under cGMP conditions that shall cover the entire 58 amino acid sequence of the TARP protein as well as include the original two 9 amino acid proteins TARP 27-35 and 29-37-9V from PolyPeptide

Laboratories 9395 Cabot Drive, San Diego, CA 92126.

This acquisition will be processed under FAR Part 12 – Acquisition for Commercial Items and will be made pursuant to the authority in FAR 13.106-1 (b)(2) and 13.501-(a)(1) using simplified acquisition procedures for commercial acquisitions. The North American Industry

Classification System code is 325414 and the employee size standard is 500.

Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract.

The delivery date is six (6) months after award of order.

It has been determined there are no opportunities to acquire green products or services for this procurement.

The NCI protocols 15-C-0075 and 15-C-0076 were approved by the NCI IRB in

January 2015; with enrollment of the first study subjects in each protocol occurring in May

2015 after Department of Transfusion Medicine personnel had been hired and trained in the manufacture of the multi-eptiope (ME) TARP peptide vaccines. As previously noted, the investigational supply of ME TARP peptides was depleted as a result of additional sterility testing associated with the PDS quarantine that began in June 2015. Consequently, enrollment in both studies has been on hold since September 2015 until a new source of vialed, cGMP TARP peptides could be identified. Completion of these studies is contingent upon manufacturing completion and delivery of the cGMP TARP peptides to the NCI

Vaccine Branch.

A. CONTRACTOR RESPONSIBILITIES

The Contractor shall produce clinical grade TARP (TCR Alternative Reading Frame Protein) peptides under cGMP conditions that shall cover the entire 58 amino acid sequence of the TARP protein as well as include the original two 9 amino acid proteins TARP 27-35 and 29-37-9V.

TARP is a novel protein that is expressed in ~90-95% of prostate and ~50% of breast cancers and is being developed as a therapeutic vaccine to treat these and possibly other cancers. The amino acid sequence of the TARP protein is below:

The use of two 9 amino acid TARP peptides (TARP 27-35 and TARP 29-37-9V) in a therapeutic vaccine platform was investigated in a pilot/phase I clinical trial conducted by the NCI (09-C-

0139/ NCT00972309) in men with stage D0 prostate cancer. The amino acid sequences of the peptides used in this first generation TARP vaccine platform are below:

The study documented that the TARP peptide vaccine was safe, very well tolerated, immunogenic and resulted in a slowing in the rate of PSA rise in the majority of subjects at 24 and 48 weeks. As a result of this success, investigators within the NCI Vaccine Branch have moved forward with the development of a second-generation multi-epitope (ME) TARP peptide vaccine platform comprised of multiple, longer, overlapping TARP peptides that they hypothesize will induce better immune responses and clinical outcomes in a broader patient population.

The overlapping cGMP peptides to be manufactured by the Contractor are 20 amino acids in length (except for the fifth peptide that is 18 amino acids in length), overlap by 10 amino acid residues and cover the entire 58 amino acid TARP protein sequence as specified below.

Specifically, the Contract shall manufacture the following seven cGMP TARP peptides using protected amino acids of non-animal origin in the quantities outlined below:

1. Peptide 1: TARP 27-35 (MPS-326) Length: 9 amino acids Amount: 1.2 gram

These new peptides are replacements for ones consumed in mandatory sterility testing after the

2015 NIH pharmacy quarantine. To maintain continuity of the clinical trial, it is essential that the replacement peptides come from the same source, PolyPeptide Laboratories since the lab cannot change products in the middle of the clinical trial. These are urgently needed to treat prostate cancer patients who are waiting to be treated.
