JUSTIFICATION & APPROVAL
Below the Simplified Acquisition Threshold ($10,000 - $250,000)
Project Title: Creating a new transgenic mouse model with pCAG-Lox-STOP-Lox-mouse Adgrf1-IREStdTomato-bGHpolyA, using a CRISPR-Cas9 method
A. Name and address of the proposed contractor(s)
The Jackson Laboratory, 610 Main Street Bar Harbor, ME USA 04609
B. Describe why only the requested supply/service will meet the government’s needs
A genetically modified mouse is planned to be used to enhance the Adgrf1 expression specifically in spatial and temporal manner to define the effect of GPR110, encoded protein from Adgrf1 mRNA, in brain function. The Jackson Laboratory was selected because the Jackson Laboratory mice are the most well-characterized and widely used mice available. The Jackson Laboratory team has expertise in designing the genetic modification of the mouse including the plasmid donor synthesis for knock in plasmid, embryo manipulation with Bxb1 editing reagents and founder identification using PCR. In this project, the Jackson Laboratory will insert pCAG-Lox-STOP-Lox-mouse Adgrf1-IREStdTomato-bGHpolyA into the C57BL/6J mouse ROSA26 locus using CRISPR/Cas embryo manipulation.
C. Describe why only the proposed contractor is capable of providing the requested supply/service
The Jackson Laboratory was the selected vendor for the following: As mentioned above, the Jackson Laboratory team trained on various model generation technologies, therefore, the right strategy will be used to make the right model. Furthermore, the colony will be expanded and preserved with the characterization in the Jackson Laboratory, therefore, the model could be safe from any case of accidental genetic modification or mutation during maintenance. The sperm cryopreservation will be a service to utilize the material for future model generation. Once the sperm cryopreservation process has been completed, the straws of sperm will be maintained in liquid nitrogen storage in three separate tanks. Two of these storage tanks are located at The Jackson Laboratory in Bar Harbor, Maine and one tank is located off-site.
