The Jackson Laboratory will generate and insert CRISPR Knock In plasmid, pCAG-Lox-STOP-Lox-mouse Adgrf1-IREStdTomato-bGHpolyA into the C57BL/6J mouse ROSA26 locus to enhance the Adgrf1 expression. This service includes plasmid design and reagent preparation (8- 10 weeks), embryo manipulation (4-5 weeks) and founder identification (4-6 weeks), CRISPR breeding service and sperm cryopreservation. The Jackson Laboratory mice are the most well-characterized and widely used mice available, and once the sperm cryopreservation process has been completed, the straws of sperm will be maintained in liquid nitrogen storage in three separate tanks at The Jackson Laboratory.
