D. Information:

1. Background

Next Generation Sequencing (NGS) data is a critical component of the research conducted by NIAMS Scientists. There has been a steady increase in the use of NGS data in research throughout the NIAMS IRP. The High-Throughput Sequencing Core Facility Manager is now processing up to 300 samples weekly on 3 Illumina Sequencers (HiSeq 2000, HiSeq2500, MiSeq), and the majority of the workflow for submitting the samples for sequencing is a manual process. The current process can no longer effectively support the number of samples submitted for processing.

Samples are submitted ad hoc. Currently there is no central repository or management of information about samples or the resulting datasets. Back in the lab, there is no insight into where in the process the samples are. All communications on status, issues, schedule, etc. are managed via email, phone and in person. Once generated, unless there is an established process within a lab, there is no way to centrally manage the data or the information about the data. For those submitting samples, the process and learning the process is inconsistent as is level of information and quality of information provided with the samples.

The scope and scale of data now being generated by NGS requires a Laboratory Information Management System (LIMS) to support NIAMS Scientists in the preparation, tracking and analysis of the data, as well as to increase the overall quality, integrity and long-term viability and availability of the data.

2. Purpose and Objectives

The purpose of this requirement will be a procurement of GenoLogics Clarity LIMS (Gold Edition) and Implementation Services to support the NIAMS Intramural Research Program (IRP) and High-Throughput Sequencing Core Facility. This product has been identified as the best possible solution, based on needs, costs and risks to provide end-to-end Next Generation Sequencing (NGS) sample tracking, data management and automated data analysis pipelines.

The objectives are to:

• Automate sample preparation, submission, scheduling and run workflow to the fullest extent possible

• Provide an online system to submit samples for processing

• Provide a consolidated repository for NGS Sample Meta-data

• Improve quality of NGS Sample meta-data provided for sequencing

• Automate initial analysis pipeline – conversion to fastq, alignment

• Provide Dashboards, Reporting, and Search capabilities
