357nm Emission: 447nm), GFP (Excitation: 482nm Emission: 524nm), Texas Red

(Excitation: 585nm Emission: 628nm)

• A universal stage must be included so that a variety of multi-well plates, flasks, or microscope slides can be visualized.

• A digital camera readout must be displayed and incorporated with the microscope.

• The system shall simultaneously accommodate 4 independent high output LED light cube illuminators with integrated hard coated fluorescence band‐pass excitation and emission filters for each cube plus a dedicated transmitted light slot.

• System must automatically recognize fluorescence cube installed and adjust software configuration accordingly.

• Software must be included for image acquisition and analysis.

• The system must simultaneously acquire images with four separate fluorescence channels plus transmitted light.

• The system must include a motorized X/Y scanning stage.

• The system must include one automated objective turret, filter set and condenser.

• The fluorescence LED illuminators must have a lifetime of no less than 50,000 hours at

100% power.

• The LED illuminators must have independent intensity control.

• The system must have a stage designed for a fully integrated onstage incubator for control of temperature, humidity and gases including CO2 and O2.

• The Z‐plane single‐step must have an accuracy of less than or equal to 0.150 μm and the random movement accuracy of less than or equal to 0.750μm.

• Instrument must include an onstage incubator which controls humidity, temperature, and gases (ex. CO2) needed for live cell microscopy and time lapse studies.

• Onstage incubator temperature range must be 22°C to 40°C.

• Onstage incubator CO2 range must be 0 to 20%.

• Onstage humidity range must be greater than or equal to 80% relative humidity at 37°C.

• The time-lapse function must have the capability to be combined with autofocus and or

Z-stack.

• The system must include a scale bar to determine size of sample.

• The system must include automated cell counting and cell confluence calculation capabilities.

• The system must include automated transfection efficiency calculation capability.

• The system must include a white LED light for transmitted illumination.

• The system must support direct output to USB and networked storage.

• The system must provide outputs in the following file formats.

- Images: jpg, tif, and png.

- Movies and time-lapses: avi and wmv.

• The system must include the ability to perform cell counting, confluence and transfection efficiency calculations of multiple images using the same analysis parameters in batch mode.

• An operations and maintenance manual (electronic, hardcopy, or both) shall be provided.

III. DELIVERY

Delivery shall occur within sixty (60) calendar days of award. Delivery shall take place during normal business hours, between 8:30 am and 5:00 pm Eastern Time, Monday through Friday except Federal Holidays, and shall be coordinated with the Contracting Officer Representative

(COR) and Subject Matter Expert (SME). Delivery shall be made to the NIST campus at 100

Bureau Drive, Gaithersburg, MD, 20899, USA.

IV. INSTALLATION

The Contractor shall install the instrument at NIST, Gaithersburg, MD. Installation shall include, at a minimum, uncrating/unpacking of all equipment, set-up and necessary connections for operation, hardware and software installation, basic functionality, instrument start-up, demonstration of instrument performance with the specifications and removal of any trash.

Installation shall take place between 8:30 AM and 5:00 PM Monday through Friday, with the exception of holidays. Installation of the system shall occur within thirty (30) days of the delivery date.

V. TRAINING

In-person training for up to two (2) people shall be coordinated through the COR and SME and shall be completed within 30 calendar days of installation Training shall include operation of the instrument and all features including camera, optics, stage, basic data analysis and file saving and exporting.

VI. WARRANTY

The Contractor shall provide, at minimum, a one (1) year warranty to include all parts, materials, labor, and travel for the instrument to keep the instrument operating to manufacturer's specifications. The warranty period shall commence upon installation of the instrument.

VII. ACCEPTANCE

The Contractor shall demonstrate that performance specifications are met and tested using NIST cell samples for compliance with the parameters of Section II. SPECIFICATIONS. Acceptance shall occur within 7 calendar days of installation.
