Section 2.
Salient Characteristics and Specifications A) Salient Characteristics of the Requested System. The requested instrument must have the following salient characteristics and specifications:
i) The Cryo System FE-SEM must use a thermionic field emission source to generate electron beam.
ii) At a minimum, the FE-SEM must be capable of delivering to the sample, accelerating voltages ranging from 0.5kV to 30kV and continuously adjustable in at least 10V steps.
iii) At a minimum, the Cryo System FE-SEM must operate at both high vacuum mode (of 5x10-4Pa) and variable pressure mode (or low vacuum mode of 7-500 Pa), to allow for investigation of conductive samples and non-conductive samples in their natural uncoated state. Switching these two modes must be controllable through the software.
vi) The Cryo System FE-SEM must be equipped with an in – Chamber E-T type Secondary Electron Detector, an in-lens Secondary Electron Detector and an in-beam Backscattered Electron Detector.
v) At a minimum, the Cryo System FE-SEM must demonstrate imaging resolution (in lens SE detector) of 1 nm at 30kV under high vacuum mode, and 1.5 nm at 3 kV under low vacuum mode.
vi) At a minimum, the FE-SEM must provide continuously adjustable current from 2pA to 200nA. The current must be adjustable through use of FE-SEM software to allow for optimizing imaging conditions under a wide variety of samples.
Vii) The Cryo System-FE-SEM specimen chamber must be equipped with active vibration isolation system to minimize vibration and to allow for high resolution imaging.
viii). At a minimum, the specimen chamber and stage of Cryo System FE-SEM must be large enough to hold samples of 4 inch in diameter.
ix). The Cryo System FE-SEM must have beam deceleration (or gentle beam) option to allow for reducing beam diameter and surface charge.
x). At a minimum, the Cryo System FE-SEM must have an Energy Dispersive X-ray Spectrometer (EDS), and the EDS system must be a Silicon Drift Detector (SDD) with detector size of 80 mm2 to allow for the detection of a limited range of X-rays emitted at low kV.
xi). The FE-SEM must provide particle analysis software that is capable of conducting automated particle morphological analysis (e.g., size, shape, aspect ratio) and chemical composition analysis. The EDS detector must be fully integrated with particle analysis software to perform chemical composition analysis.
xii). The Cryo System FE-SEM must be capable of observing non-stained and untreated vitreous samples.
xiii). The Cryo System of FE-SEM must be a modular design that is capable of minimizing biosafety concerns of biological samples and protecting the samples from contamination.
xiv). The Cryo System of FE-SEM must consist of Cryo System Transfer Unit, Freeze Fracture Unit and Cryo System Coater Unit. The Cryo System Coater must be configured for versatile functions such as sputtering, carbon evaporation and e-beam evaporation in both room-temperature and Cryo System Condition.
xv). The decking unit of Cryo System on the FE-SEM must be geometrically minimized to maximize the installation capability of other SEM features (e.g., EDS) and to allow for any future upgrades.
xvi). The sample holder and decking unit design of the Cryo System must generate minimum vibration to maintain the resolution of FE-SEM.
xvii). The FE-SEM must come with computers and software to operate the microscope with full function.
B) Service Agreement
The FDA requires an Annual Preventative Maintenance (PM) Service coverage on the Cryo-Field Emission Scanning Electron Microscopy (Cryo FE-SEM) with an Energy Dispersive X-ray Spectrometer (EDS):
The requested service agreement must include labor, travel, living expenses and spare parts coverage.
The service must include two (2) annual Preventative Maintenance (PM) visits for SEM, one (1) annual Preventative Maintenance visit for EDS and Cryo System at periodically, mutually-agreed times.
The preventative service must be provided by “manufacturer trained and qualified” field service engineers.
The service must cover the system’s software and/or software updates applicable to customer’s current configuration to maintain full performance of the instruments.
Service coverage must be available during normal business hours (Monday through Friday) with a targeted response time of forty-eight (48) hours for on-site labor and next business day delivery of spare parts.
The contractor must provide unlimited technical support via email, phone or web.
C) Other Considerations.
i). The contractor shall provide proof of sales of FE-SEM Systems that are integrated with a similar Cryo System and/or EDS Detector, by listing two (2) contracts completed during the past five (5) years, similar in size and scope to the requirements outlined in the Statement of Work. Offerors shall include the following information for each contract identified: Name of Contracting Organization, Contract Number, Contract Type, Total Contract Value, Description of Requirement and Specific Responsibilities of the Offeror (as they relate to the salient characteristics and specifications provided in this RFQ) and Contract Period of Performance.
ii). The contractor shall provide all personnel and equipment necessary to deliver and install the FE-SEM System at the FDA Delivery location in section 3.18.2. Furthermore, the contractor shall provide an application’s specialist and service engineer to perform resolution imaging tests for comparison to factory image tests following delivery.
