About this file

This document is a purchase description for a federal solicitation. The National Institute of Neurological Disorders and Stroke within the National Institutes of Health requires a Becton, Dickinson & Company BD FACSCelesta flow cytometer analyzer system or equal. The system will be used by principal investigators and scientific staff to conduct cell sorting, cell function analysis, and multi-parametric analysis of single cells to further research goals related to glioblastoma and glioma stem cells. The system must have three lasers with 14 parameters and 12 colors, fiber optic light delivery, a polygon optical arrangement with three pinholes for up to three lasers, easily replaceable optical filters and mirrors, and sensitivity for detecting fluorescent signals from single cells. The related federal contract opportunity is solicitation number 75N95023Q00662 for this flow cytometer system from the Department of Health and Human Services National Institutes of Health National Institute on Drug Abuse.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

Title: Becton, Dickinson & Company BD FACSCelesta™ or Equal

1. Statement of Need and Purpose:

The purpose of this requirement is to procure a quantity of one (1) Becton, Dickinson & Company BD FACSCelesta™ or equal flow cytometer analyzer system for the Neurosurgical Oncology Unit (NOU) within the National Institute of Neurological Disorders and Stroke (NINDS), Division of Intramural Research (DIR). A flow cytometer analyzer system is needed for cell sorting and cell function analysis in order for NINDS principal investigators (PIs) and scientific staff to continue its ongoing research critical to its NINDS Mission.

2. Background Information and Objective:

The NINDS, which is an Institute at the National Institutes of Health (NIH), mission is to seek fundamental knowledge about the brain and nervous system and to use that knowledge to reduce the burden of neurological disease for all people.

The NOU aims to develop novel therapeutics for Glioblastoma (GBM)–the most common primary malignant CNS. Despite aggressive management with maximal surgical resection, radiation, and chemotherapy, median and 5-year survival rates remain low (15 months and 5.6%, respectively). Tumor-specific contributors to treatment failure include cellular and genetic heterogeneity and a highly immunosuppressive tumor microenvironment. GBMs also harbor self-renewing, tumorigenic glioma stem-like cells (GSCs) that contribute to tumor initiation, maintenance, and recurrence. The need for novel GBM therapeutic strategies is urgent.

The NOU uses several molecular biology and biochemistry techniques in its scientific mission to better understand pathogenesis of gliomas and develop better therapies.

While the Unit performs several assays such as cell cycle, it does not have a technology that provides rapid multi-parametric analysis of single cells in soluti , etc., to efficiently and reliably do this work.

Therefore, the BD FACSCelesta™ is needed for the NOU to perform several assays in a single unit and provide intra-laboratory high content flow cytometry for the laboratory that will be compatible with current research protocols within the NIH.

3. Generic Name of Product:

Flow Cytometer Analyzer System

4. Purchase Description:

For reference purposes, the brand-name system that meets the need of this requirement is as follows:

(1) Specify trade name: FACSCelesta Standard

(2) Specify product’s brand name: BD FACSCelesta™

(3) Specify manufacturer’s name: Becton, Dickinson & Company

(4) Part number(s): FACSCelesta Standard

5. Salient characteristics:

The significant, essential physical, functional, performance, or material element(s) that are the Government’s minimum acceptable standards for an equivalent system to meet the government’s objective are as follows.

1. Must have 3 lasers, which has 14 parameters with 12 colors.

2. Must have state of the art fiber optics to carry the light/fluorescent signal from the flow cell for the PMTs.

3. Must have a polygon optical arrangement and 3 pinholes for up to 3 lasers.

4. Must have easily replaceable optical filters and mirrors for experimental flexibility.

5. Must have sensitivity of <30 MESF-PE, <80 MESF-FITC and <75 MESF-APC that can perform real-time statistics with software, which must have the CS and T software for standardizing and QC the instrument (such as Cytometer Setup and Tracking software).

6. Must fully automate the QC of numerous parameters and alerts the user if a parameter does not meet specification and tracks the QC parameters over time.

7. Must have capability of analyzing at least 30 logical gates.

8. Must have digital electronics with acquisition speeds of 25000 events per second.

9. Must use continuous flow of sheath fluid, no syringe-based systems, for looking at unlimited data volume or rare event analysis.

10. Must have up to 20-liter capacity for sheath fluid and waste thus allowing the user to run the unit for long periods of time without needing to add Sheath fluid doing long runs may be added post installation.

11. Service technicians and engineers must be BSL-3 and Original Equipment Manufacturer (OEM) certified for its on-site installation, set-up and training of the equipment.

12. Extended warranty for all electronic, optical and mechanical components will be free from defects shall be at least five (5) year from the date of installation and inspection and acceptance by the Government. Warranty service shall include trouble-shooting capabilities based on complete knowledge of the entire system.

Warranty shall not include routine or periodic maintenance services.
