STATEMENT OF WORK/REQUIREMENTS DOCUMENT

Title: Supercontinuum light source with filters Requesting Lab: PML

I. BACKGROUND INFORMATION

The gene therapy manufacturing innovations in measurement sciences project goal is to develop light scattering microscopy methods to characterize gene delivery particles. As part of that work, we are building multispectral light scattering microscopes and a library of the refractive indices of gene delivery particles and their subcomponents.
A new, high-power, broadband VIS-NIR supercontinuum light source is required for the light scattering microscopes and for the refractometer that will be used for the refractive index library measurements. Two different tunable, spectral filters for the light source are also needed to select specific wavelength bands to measure the dispersion in the refractive index and scattering properties of gene delivery particles. The measured dispersion will be used to quantify the composition of each particle. The supercontinuum light source and filters will be shared between the microscopy and refractometry apparatus. One spectral filter is needed for microscopy measurements with a wide and tunable spectral bandwidth for investigating how the light source bandwidth affects measurement accuracy. A separate tunable, multiline spectral filter with narrow bandwidths and fast wavelength switching time is needed for refractive index measurements with high spectral resolution, and for performing fast wavelength switching while recording microscopy videos of gene delivery particles.

Purpose/Objective: The objective of the gene therapy manufacturing project is to develop new methods to better characterize gene therapy particles at high throughputs. Emerging gene therapy treatments use gene delivery nanoparticles, such as viral vectors and non-viral carriers, to cure intractable diseases. These manufactured particles suffer heterogeneity issues that hinder treatment efficacy and safety. This project aims to develop light-scattering microscopy methods to enable rapid, multi-attribute measurements of individual gene therapy nanoparticles’ physical characteristics. Such measurements will speed the development of new gene therapy treatments and improve their efficacy and safety. The supercontinuum light source and tunable filters are required to meet this objective.
