This document describes a purchase description for a confocal laser scanning microscope system and multi-photon scanning system. The National Institutes of Health, National Institute on Drug Abuse is seeking to procure one microscope system and one multi-photon scanning system to be used for imaging Drosophila brain and tissue stained with multiple probes and fluorophores. The confocal system must be able to heavily multiplex different fluorophores on the same tissue sample and take high-quality images. The multi-photon system will be used for live imaging of Drosophila brain. Key requirements for the systems include specific laser wavelengths, scan speeds, image formats, and software for acquisition and analysis. The vendor must provide installation, setup, and initial training. Equipment must be delivered within 16 weeks to Building 35 at NIH in Bethesda, Maryland between 8am-5pm EST Monday through Friday. The period of performance includes at least one year of warranty on the systems.

The related federal contract opportunity is solicitation number 75N95023Q00334 for ZEISS LSM900 AND 980 SYSTEMS OR EQUAL through the Department of Health and Human Services National Institutes of Health National Institute on Drug Abuse.

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

Title: Zeiss LSM900 and LSM980 system or Equal

1. Statement of Need and Purpose: The purpose of this acquisition is to procure laser point-scanning confocal microscope systems designed for imaging stained and live tissue. The confocal laser microscope will be used to image Drosophila brain and other tissue stained with multiple antibodies with/or fluorescent in situ probes. The primary purpose is to be able to heavily multiplex different fluorophores on the same brain/ tissue and take high-quality images to find out dendrite morphology change. The multi-photon scanning system will be used to take live imaging of Drosophila brain with a fluorophore probe. With our current multi-photon laser turned to 920nm, the system has a wide range of detection, which can simultaneously scan and collect fluorophore movement in the live brain. The collected data will be used for the analysis of dynamic change at the early dendrite development stage.

2. Background Information and Objective:

Dendrite Morphogenesis and Plasticity Section( DMPU) is a section of NIH/NINDS.

Applying the Drosophila larval visual circuit, DMPU investigates the structural and functional plasticity change to find the mechanisms underlying the neurolonal development. We need to perform large-scale genetic screens on drosophila with a confocal system to take high-quality images to determine the dendrite morphology change at the cell level. Furthermore, analyze these candidate genes using in vivo live imaging with a multi-photon scanning system. The primary approach is understanding the molecular path to regulate dendrite morphogenesis and structural plasticity and learning the consequent function change due to deficits in homeostatic neuronal plasticity. The confocal laser microscope and multi-photon scanning system described above will allow the DMPU to screen large-scale genetically modified drosophila containing fluorescent reporters to learn certain gene functions and lively tracing fluorescent signal movement in the larva brain.
