About this file

This document includes a purchase description and related federal contract opportunity solicitation. The purchase description seeks fluorescent in situ hybridization probes that can detect and quantify specific RNA molecules in mouse brain tissue. The probes must identify 40 genes without cross-reactivity and provide quantitative single-molecule results. They must also multiplex the detection of 40 genes in the same tissue sample. The quantity needed is 40 probes, to be delivered within one week to the National Institutes of Health. The related solicitation is for the RNAScope HiPlex Probes and Reagents from Advanced Cell Diagnostics on a non-competitive basis under simplified acquisition procedures. The solicitation number is 75N95024Q00153 and is issued by the National Institute on Drug Abuse on behalf of the National Institute of Neurological Disorders and Stroke.

PURCHASE DESCRIPTION

1. Statement of Need and Purpose: The Talmage laboratory at the National Institute of

Neurological Disorders and Stroke (NINDS) seeks to purchase probes to identify and quantify RNA molecules belonging to specific genes. These probes are to be used in mouse brain tissue to validate and enhance findings from previous studies that the lab has been undertaking. The chosen method for this is the RNAScope assay as it allows the ability to detect 48 genes in the same tissue in a quantitative manner. The lab has previously used this technology and has purchased the equipment necessary to perform this assay.

2. Background Information and Objective: This project seeks to understand how genetic susceptibility to schizophrenia due to a mutation in the Nrg1 gene influences brain development and function. In relation to this, we identified that mice carrying a mutation in the Nrg1 gene showed altered gene expression, neuronal morphology, and altered fate of neural stem cells in the hippocampus. Further analysis has led us to identify overabundance of a rare and poorly characterized cell type in the dentate gyrus.

Using single nucleus transcriptomics we have identified several new potential marker genes for these cells which we now need to validate empirically and quantitatively.

3. Generic Name of Product: Fluorescent insitu hybridization probes to detect RNA molecules with a capacity for multiplexing.

4. Purchase Description: RNAScope HiPlex Probes and Reagents (Brand Name Only)

5. Salient characteristics: The RNA Insitu hybridization probes must be able to specifically detect the genes and specific gene isoforms without cross reactivity. The product should also be able to provide quantitative results at the single molecule level.

Additionally, it should have the ability to multiplex the detection of 40 genes in the same tissue sample to ascertain the co-expression (or lack thereof) of these genes in the same cells.

6. Quantity: 40 probes

7. Delivery Date: Within 1 week after Receipt of Order

8. Delivery Address: 35 Convent Dr.

Bldg. 35 Room 3B-1010

National Institutes of Health

Bethesda MD 20892
