Version 11-30-2016-1 FAR Part 11—Describing Agency Needs

PURCHASE DESCRIPTION

Total Internal Reflection Flouroscent Microscope System (CoSMoS, MicroMirror TIRF Microscope)

Dr. Joe Mindell (MTBU), NINDS

1. Statement of Need and Purpose:

The Membrane Transport Biophysics Unit (MTBU), National Institute of Neurological Disorders and Stroke (NINDS) needs to use cutting-edge imaging approaches to investigate the functioning of lysosomes and related transporter activity. The laboratory requires a TIRF (total internal reflection fluorescence) microscope to perform the research.

2. Background Information and Objective:

The MTBU performs experiments examining the function of lysosomes from acidification and ion fluxes to cell metabolism and amino acid equilibrium. Lysosomes play a key role in the metabolism of the cell and have been implicated in numerous lysosomal storage disorders as well as broader systemic diseases such as cancer and diabetes. Recent technological advances have enabled the imaging of individual lysosomes, who's function can vary across the distribution of lysosomes in a cell which is obscured in ensemble assays. The microscope will enable lab personnel to measure the ion fluxes and amino acid homeostasis in individual, native lysosomes as well as study, in isolation, the transporters that perform these key roles which have been implicated directly in numerous diseases.

3. Generic Name of Product:

Total Internal Reflection Fluorescent Microscope System

4. Salient characteristics:

The contractor shall provide the NINDS MTBU with the hardware and software brand name or equal to The RM21® MicroMirror TIRF microscopy system from Mad City Labs, Inc.

• Enables implementation of the TIRF imaging method. The system shall be useful for low-signal, multi-wavelength TIRF applications in basic biological research.

Can be used for single molecule approaches to study protein domain orientations and conformations, and experiments addressing the assembly and activity of multi-component biomolecular machines.

• Shall be able to accommodate sample positioning and optical pathway space constraints, and full and open access to both entrance pupil and back aperture.

• The lens shall be statically mounted directly to the base plate of either a manual or motorized x-y micropositioning stage, with 25mm of travel along each axis.

Stage shall be able to hold a fully integrated 3-axis nanopositioning stage. The nanopositioner (LP200) shall have 200 microns of travel in each dimension. A

Version 11-30-2016-1 FAR Part 11—Describing Agency Needs manual Z-axis micropositioner (12mm travel), shall include an integrated sample holder, and be mounted directly on top of the nanopositioner so that it moves in all 3 dimensions with the nanopositioning stage.

• The sample positioning assembly shall be integrated with an optical microscopy platform providing space for, and access to the excitation and emission light pathways. The excitation beams shall be directed into and out of the objective lens using two cylindrical 45o micro-mirrors that are positioned at opposite edges of the exit aperture of the objective lens. The micro-mirror mount shall incorporate 6 axes of positioning to facilitate adjustment of the micro-mirrors.

• The emission TIR beam shall be monitored by a TIRF-Lock system to ensure TIR signal is maintained. The TIRF-Lock system shall include a sensor, sensor electronics and software.

• The entire TIRF microscopy system shall be controllable via software provided.

5. Delivery Date:

The Contractor shall deliver and install the equipment within 90 days after receipt of order. Delivery must be Freight on Board (FOB) Destination and include inside delivery.

The equipment shall be delivered and installed in coordination with the Contracting Officer’s Representative (COR). The contractor shall provide an Original Equipment Manufacturer (OEM) certified service technician for setup, installation and training.

6. Other Requirements:

A warranty for a period of at least 1 year shall be provided following the date of delivery. All products shall be warranted to be free from material defects in workmanship or material under normal use and service for the warranty period. The contractor shall provide software upgrades during the warranty period. Periodic preventative maintenance visits cannot be included in warranty.
