Statement of Work

Development of Transgenic Commercial Citrus Tolerant to the Citrus Greening Pathogen, Candidatus Liberibacter asiaticus

Background

The United States Department of Agriculture - Center for Plant Health Science & Technology (CPHST), is the scientific support agency of the Animal Plant Health Inspection Service, Plant Protection and Quarantine division. Plant Protection and Quarantine is responsible for safeguarding American agriculture from exotic pests and diseases and to provide response to incursions by eradication or containment.

The high consequence exotic Citrus Greening pathogen, Candidatus Liberibacter asiaticus, was discovered in Florida 2005 and is now established throughout the major citrus growing regions of the state. Through the Citrus Health Response Plan, CPHST leads the effort to limit spread of this pathogen within Florida and to other citrus growing states where the pathogen is not present.

There is no known source of viable resistance in Citrus species to Citrus Greening, and pesticides to the pathogen or to its psyllid vector is not effective in disease prevention.

Citrus growing regions that have high prevalence of Citrus Greening become unproductive after a few years and this is anticipated to occur in Florida as disease prevalence increases. The future viability of the Florida citrus industry is dependent on finding a suitable means of production in the presence of the Citrus Greening Pathogen.

It is imperative that a durable means of growing citrus that remains productive long enough for sustainable harvest be found. At this time, the development of resistant citrus germplasm through genetic engineering provides the most likely means of achieving this goal.

Objective

To produce within an 18 month window mature Valencia and Hamlin orange tissue germplasm transformed (genetically modified) to express an array of genes designed to impede growth of pathogenic bacteria specifically to provide demonstrated resistance to the Citrus Greening pathogen present in Florida commercial citrus groves.

Work to Be Performed

The Principle Investigator of the contract laboratory will furnish services and advice to the United States Department of Agriculture - Center for Plant Health Science & Technology located at 1730 Varsity Drive, Suite 400, Raleigh, NC 27606. Services will include the following:

1. Provide experimental results on methods developed to transform Valencia and Hamlin orange tissue with constructs suitable for addition of genes that provide antimicrobial properties to these Citrus species.

2. Provide and make available any transformed Citrus explants under the purview of this project for additional testing and development.

3. Provide experimental results on transformation of Valencia and/or Hamlin orange tissues with genes that provide antimicrobial properties including incorporation and placement of genes and their stability, as well as transcript and protein expression and stability.

4. Provide experimental results on effectiveness of transformed Valencia and/or Hamlin orange tissues with genes that provide antimicrobial properties to challenges of inoculation with the Citrus Greening pathogen, Candidatus Liberibacter asiaticus, by dodder, grafting, and or vector transmission.

5. Provide and make available transformed Valencia and/or Hamlin orange explants demonstrated to provide antimicrobial properties.

6. USDA CPHST retains all rights to transformed lines developed under this contract.

Time Line

Work is to begin immediately upon issue of contract and completed within 24 months with the CPHST objective of generating disease resistant Citrus germplasm.

Milestones and Payment Schedule

1. Review and Action Plan – Contract Start + 1 Month

Review contract requirements and prepare action plan brief for CPHST review.

2. Initial Experiment and Status Report – Contract Start + 6 months

Prepare brief report on status of transformed lines and genetic construct development.

3. Transformation of Citrus tissue – Contract Start + 1 year

Report on genetic characteristics of transformed lines and potential for antimicrobial testing. Forwarding of promising lines for further method development.

4. Report of ‘proof of concept’ strategy for genes with antimicrobial activities –

Contract Start + 18 months

5. Final report on microbial testing and forwarding of explant lines of mature tissue for further method development – Contract Start + 24 months

An invoice for a progress payment based on hours expended is to be submitted every 180 days and accompanied by a progress report and other deliverables as identified in the above Milestones section. Payment will be made upon satisfactory review of progress.

Additional Contract and Proposal Information

This is a firm fixed price level of effort contract therefore the contract fee shall include all consultant fees, travel cost and meal and lodging costs for the Principle Investigator and/or laboratory personnel.

Variation of Milestones

Any variation to milestones will be by mutual agreement between CPHST and the contractor, and will be made in writing.

Qualifications

The Government will award a contract resulting from this solicitation to the responsible offeror whose offer conforming to the solicitation will be most advantageous to the Government, price and other factors considered. The following factors shall be used to evaluate offers (see evaluation factors below):

Technical Capability Past Performance

Technical and past performance, when combined, are more important than price. Please note that a failure to meet any of the technical capabilities may disqualify a proposal from award. The Government intends to evaluate proposals and make an award without discussions but reserves the right to conduct discussions if necessary.

Technical Capability Evaluation Factors:

The minimum acceptable scientific laboratory qualifications are as follows:

1) Experience at the transformation of a variety of plant hosts with suitable ‘freedom-to-operate’ for engineering technologies for Citrus species

a) Development of transformation technologies of physiologically mature Citrus

2) Demonstrated ability to manipulate genes with antimicrobial properties:

a) Suitable ‘freedom-to-operate’ a variety of genes with demonstrated antimicrobial activities

b) Demonstration of experience and data that quantify parameters of antimicrobial efficacy of system

c) Ability to test and identify high resistance to Citrus Greening pathogen in Citrus hosts

3) Facilities suitable for Select Agent research and regulatory permits in place for experimentation with Citrus Greening pathogen Candidatus Liberibacter asiaticus.

Submittals

Proposals must include, at a minimum, enough information to evaluate each of the above items and shall include a resume or curriculum vitae of the principal researcher(s).

Past performance information shall include 3 – 5 references on research projects or contracts of a similar nature.
