About this file
This document is a single source justification for the acquisition of an ABI QuantaStudio Digital PCR System from Applied Biosystems. The Fish and Wildlife Service Bozeman Fish Health Center requires the system to provide absolute DNA quantification with the simplicity of other ABI qPCR systems already in use. The single source is justified because the ABI system utilizes a proprietary microfluidic array plate technology that enables all digital PCR steps to be conducted on one instrument, minimizing wasted sample volume compared to other platforms. It is also compatible with the assays and software used by the other ABI PCR machines at the Center, allowing for easier data sharing and comparison across instruments. The anticipated cost is considered fair and reasonable based on similar equipment prices. The justification was approved by the program officer on June 27, 2023 and the contracting officer on July 17, 2023.

1. Description of Need
3. Other Capable Sources
2. Market Research Conducted
Pursuant to FAR subpart 13.106-1(b)(1)(i) and (ii), for purchases not exceeding the Simplified Acquisition Threshold (SAT), Contracting Officers may solicit from only one source if the circumstances deem only one source is reasonably available (e.g., urgency, exclusive licensing agreements, brand name, or industrial mobilization).
Instructions: The information in the text boxes are provided as sample language or to indicate a needed fill-in. Sample language should be removed from the final document. All text boxes require information and shall not be left blank.

| Text3: Other sources of Digital PCR machines do not utilize a plate-based digital PCR platform powered by proprietary microfluidic array plate (MAP) technology which enables all the necessary steps for dPCR—compartmentalizing, thermal cycling, and data acquisition—to be conducted on a single instrument. The technology of this particular machine minimizes wasted sample volume by leveraging the MAP technology so over 95% of the input sample is analyzed per reaction compared to the 25–60% of other dPCR platforms. |

Other Digital PCR systems do not use the same or even similar software that machines already purchased and in use at the Bozeman FHC use making data sharing/comparison across machines difficult or non-existent.
Other systems do not utilize the same large portfolio of TaqMan assays already in use at Bozeman FHC.
| Text1: This ABI Digital PCR system provides absolute DNA quantification with the simplicity of other ABI qPCR systems and has the same data format and qPCR-like workflow as the several other ABI PCR systems already at and used extensively by the Bozeman FHC. This ABI machine will result in easier data sharing and data comparison between machines, and enhanced compatibility between all ABI PCR machines at Bozeman FHC. Compatible with other assays already used at Bozeman FHC for gene expression, genetic variation, gene regulation, or protein expression experiments. | |
| Text4: Other machines with digital PCR capability were researched and competitor systems do not allow the amount of flexibility of going from one to the other on the same machine. Further, competitors do not utilize the proprietary plate technology which enhances efficiency and saves on wasted sample volume and time. | |
| Text6: Anticipated cost is considered fair and reasonable due to comparison of proposed costs/prices with costs/prices for similar supplies or services. | |

Text5: The Bozeman FHC already owns several ABI PCR systems and all have had no mechanical issues after many years of use. This contractor provides results we can trust and has improved well-to-well and instrument-to-instrument data accuracy systems.
