STATEMENT OF WORK

Closed vessel microwave digestion of low mass biologic samples for trace element analysis

The Department of the Interior (DOI), U.S. Geological Survey (USGS) Wyoming – Montana Water Science Center (WY - MT WSC), 3162 Bozeman Avenue, Helena, MT 59601 collects macroinvertebrate, algae, and fish samples from streams, lakes, and laboratory experimentation in support of multiple projects studying the effects and transfer of metals in aquatic systems. Biological samples are frozen immediately after collection by the USGS. They are then freeze dried, weighed, and sent to a contractor for analysis.

SCOPE

USGS WY-MT WSC requires a facility to perform closed vessel digestion of low mass biological samples for trace element analysis. Specifically, 57 element ICP-MS following Wolf and Adams (2015) methods to maintain data quality. The USGS will provide the contractor freeze dried biological samples. The contractor will provide all equipment, labor, travel, shipping, and materials for digestion and analysis.

TECHNICAL REQUIREMENTS

1. The contractor will analyze low mass biological extract sample for 57 elements (Table 1) using ICP- MS following Wolf and Adams (2015) methods to maintain data quality.

Table 1. 57 elements included in analysis Ag Ca Cu Gd Li Nb Rb SO4 Tl Yb Al Cd Dy Ge Lu Nd Sb Sr Tm Zn As Ce Er Hf Mg Ni Sc Ta U Zr Ba Co Eu Ho Mn P Se Tb V Be Cr Fe K Mo Pb SiO2 Th W Bi Cs Ga La Na Pr Sm Ti Y

2. The contractor must have a micro-balance and the ability to work with samples that are low in mass (2

– 5 mg).

3. A microwave (CEM MARS 6 or equivalent) to do closed vessel digestion using ultrapure nitric acid and hydrogen peroxide. Samples are then diluted with deionized water to achieve a final concentration of 20%. Sample masses can range from 0.5mg-50 mg. Final volume after dilution is typically 10mL.

(Note: there are occasionally small projects that utilize small masses and therefore the digestion volume is typically 2 mL. For those samples only, we will request a subset of 5-7 elements for analysis).

4. For digested solid samples, the sample dry weights and volumes of reagents used will be retained by the contractor. Additionally, the names of the Standard Reference Materials (SRMs) will be retained by the contractor to facilitate % recovery calculations based on certified values for a given element.

The contractor will retain any remaining sample until the USGS has received and evaluated the data.

After that, the remaining sample shall be returned to USGS.

5. Turnaround time should be less than 6 months which would include time of delivery of samples to contractor to delivery of results to USGS.

6. The contractor is responsible for Quality Control (QC) measures that are within the USGS standards as described in this Statement of Work.

DELIVERABLES

The Contractor will arrange and pay for shipping (including hazardous material shipping) and packaging both to and from the USGS, if necessary.

The Contactor shall deliver in electronic form as a spreadsheet (.xls) to the USGS containing results for all samples, blanks, and QC samples. The spreadsheet should also contain: date analyzed, detection limits

140G0322Q0145 Page 4 of 23

(clearly identified as MDL, LOQ, LOD, etc), calculated % recoveries for SRMs, and a summary of data normalization. The file should contain metadata to include samples used for the pooled QC and any modifications made to the sample (e.g. filtered, diluted). The contractor must provide follow-up technical communication to the USGS concerning results by phone or email.

MINIMUM QUALITY CONTROL (QC) MEASURES INCLUDED BY THE CONTRACTOR

Before running unknown samples, the calibration is verified using one or more independent calibration verification (ICV) standards that are prepared from a stock originating from a different supplier than the stock(s) used to prepare the calibration standards. The ICV solution should be prepared at a level near the mid-point of each the calibration curve for each element. To verify calibrations for all 57 elements present in the calibration solutions, three separate ICV solutions are typically used—see table 7. The measured ICV concentrations must be within ±10 percent of the true or prepared value. If the ICV results are not within acceptable limits, then the source of the error should be identified and corrected and the instrument recalibrated and the ICV solution(s) reanalyzed. Common errors include improperly prepared calibration standards or stock solutions and inadequate mixing of the internal standard solution with the sample solutions. The results for all quality control sample (CRM or SRS) analyses should be within ±20 percent of the certified or proposed values, including stated errors. If the results are not within ±20 percent of the stated values, the samples are prepared again and reanalyzed by ICP-MS. If the results are still not within acceptable values, the instrument and method parameters are checked and the instrument is recalibrated using freshly prepared calibration standards and the quality control samples are reanalyzed. For every 10 unknown samples, a blank, continuous calibration verification (CCV), and a quality control sample are analyzed. The CCV solution(s) are prepared from the same stock standards used to prepare the calibration standards and may be one or more of the calibration standards reanalyzed to verify continuing calibration accuracy. The measured CCV concentrations should be within ±10 percent of the true or prepared value. If the CCV values are not within limits, the instrument and method parameters are checked and the instrument is recalibrated using freshly prepared calibration standards and any affected quality control and unknown samples are reanalyzed. If the quality control sample is not within ±20 percent of the accepted value, the instrument and method parameters are checked and the instrument is recalibrated using freshly prepared calibration standards, and any affected quality control and unknown samples are reanalyzed.

The instrument detection limits and reporting limits are calculated from blanks analyses. The preferred masses for reporting are selected based on background equivalent concentration (BEC), achieved detection limits, analytical performance of quality control samples, and minimal interferences at the particular mass for a particular sample matrix. For some elements, more than one mass is analyzed in order to rule out possible interferences in unusual samples. Prior to reporting, the results for Si and S are converted to SiO2 and SO4 using 2.139 and 2.995 conversion factors, respectively. The method detection limit (MDL) is calculated as a value equaling three times the standard deviation (SD) of a large number (n > 40) of blank analyses during a 3-month period. The lower reporting limit is established by rounding the calculated MDL to a suitable number of significant figures and raising reporting levels for ubiquitous elements based on professional judgment to prevent false positives. The instrument MDLs and reporting limits are reevaluated on an annual basis or as instrument operating parameters are changed. The upper reporting limit is generally extended to two times the highest calibration standard. For samples with results reading above the highest calibration standard, either a quality control sample at a higher level or a high-level calibration standard can be used to verify linearity above the highest calibration concentration.

The 6 quality control samples utilized for repeat analysis and the ICV samples are used to check the method performance during analyses of unknown samples. The accepted or most probable value (MPV) for element concentrations in the USGS Standard Reference Sample (SRS) waters are determined via multilaboratory round-robin analysis. For many elements in the USGS SRSs, the levels are near the detection limits and are useful for evaluating accuracy at low concentrations.

QUALITY ASSUANCE (QA) PROTOCOLS TAKEN BY THE USGS

The USGS is responsible for Quality Assurance. The USGS will review all sample data, quality control data, and blanks to ensure results are within the expected range. Reanalysis of samples flagged as aberrant

140G0322Q0145 Page 5 of 23 may be requested by the USGS at the contractors’ expenses.

Evaluation of Quotations and Submission Instructions

A. Overview. The Government may perform a comparative evaluation (comparing offers to each other) to select the contractor that is best suited and provides the best value, considering the evaluation factors in this solicitation. The following factors will be used to evaluate quotations:

• Firm experience and key personnel

• Technical approach

• Past performance

• Price

(1) Non-Price Factors:

(a) Experience. Experience is the extent to which a prospective contractor has performed similar work before. Recent and relevant experience is preferred. Experience with private and federal clients is preferred, as well as demonstrated experience processing permits for similar projects. In evaluating experience, we will evaluate firm’s experience as well as experience of key personnel proposed to be assigned to this project, as applicable. Individuals named in your quote which tie to the experience evaluation, must be assigned to work on the project, if awarded; and quotations must specifically affirm this requirement.

(b) Technical Approach. Regarding technical approach, we request that the prospective contractors describe the scope of their services and how you intend to fulfill the USGS’ stated objectives and SOW.

Essentially, how do you propose to approach the work, plan the work, do the work, track the work, etc., and produce the results?

(c) Past performance. Past Performance is the measure of how well the prospective contractor has performed similar work before. Recent and relevant past performance is preferred.

(2) Price Factor:

(a) Regarding price, it will be a firm-fixed price award.

B. Format for Submission of Quotations. It is the prospective contractor’s responsibility to submit sufficient information to allow a meaningful evaluation in each of these areas. Failure to do so will negatively impact our evaluation of your quotation. At minimum, we suggest you submit:

• Information concerning your firm’s experience, specifically discussing recent and relevant similar jobs completed, and describe how this is relevant to the current requirement and stated evaluation factors noted above;

• Information concerning applicable, subcontractors’ corporate experience (if subcontractors are to be used);

• Information concerning the experience of key personnel who would be working on this project;

• Information concerning previous customers or clients’ evaluation of your recent and relevant performance, such as named references, contract numbers and amounts, and point of contact information (phone/email) as applicable. We may utilize your provided references; government past performance databases; and reserve the right to use any other source of past performance or customer references information available.

• Narrative technical approach document. Please limit this narrative to no more than ten (10) single-sided pages with normal size font and spacing (i.e. not less than 10 point, not less than single spacing with normal paragraph, line, and page breaks).

C. Comparative Evaluation/Best-Value Selection. The evaluation will result in an evaluated price for each quotation (price factor), and an evaluated “most advantageous” quotation for each of the non-price factors (non-price factors). Subsequently we will perform a comparative evaluation between the price and non-price factors to determine the offer that is most advantageous to the government for award.

140G0322Q0145 Page 9 of 23

Note that our comparative evaluation and judgement as to which quotation offers the best value to the government, price and non-price factors considered, is necessarily subjective in nature. It should also be noted that the comparative evaluation may result in award to a superior technical proposal that is not the lowest price.
