About this file
This is a Statement of Work (SOW) for the procurement and maintenance of an Illumina DRAGEN Server v4 for bioinformatics data analysis of next-generation sequencing (NGS) data at the National Cancer Institute's Laboratory of Pathology. The NCI-COMPASS program currently operates TSO500 v1 analysis using a local app installed on the NIH Biowulf cluster. With Illumina's discontinuation of TSO500 v1 and transition to TSO500 v2 (which requires the DRAGEN server), NCI is procuring a second DRAGEN server to support approximately 1,000 clinical cases annually. The primary objective is to acquire the DRAGEN Server v4 (Catalog Number: 20032797) with pre-installed Dragen TruSight Oncology 500 and TSO500 ctDNA analysis software, along with comprehensive maintenance and support services. Delivery is required within 30 days of purchase order award to NIH, NCI, 10 Center Drive, Building 10, Room 3S235A, Bethesda, Maryland 20892.
The contractor must provide all hardware, labor, materials, and equipment necessary for installation, maintenance, emergency repair, and software updates. Key requirements include on-site and emergency services during standard business hours (8:00 AM–5:00 PM EST, Monday–Friday) and extended hours (5:00 PM–9:00 PM EST) with a four-hour initial response and 72-hour on-site service response guarantee. The contractor shall provide replacement parts, defective software replacement, unlimited clinical technical telephone support (8:00 AM–9:00 PM, Monday–Friday), and qualified factory-trained personnel with backup support. A 12-month manufacturer warranty, 12 months of technical support, and one year of Advance Exchange support are included in the initial year. The contract is structured as a Firm-Fixed-Price Purchase Order with quarterly payment in arrears. All costs including shipping, handling, and installation charges must be included in the quote. The contractor shall submit invoices electronically to the Treasury's Invoice Processing Platform with copies to the Technical Point of Contact and Contracting Officer. Deliverables include hardware delivery reports, emergency maintenance service reports, and software update reports due within specified timeframes in Word or PDF format. Standard federal acquisition regulations and clauses apply, including those related to small business reporting, equal employment opportunity, anti-trafficking, and environmental compliance.

STATEMENT OF WORK
1.0 TITLE
Bioinformatics data analysis of next generation sequencing (NGS) data from on-premise Illumina DRAGEN Server v4.
2.0 BACKGROUND
The NCI-COMPASS program at the Laboratory of Pathology has been running TSO500 v1 since 2019. This panel consist of 523 cancer specific genes, and it is optimized to provide high sensitivity and specificity for low-frequency somatic variants across these genes. DNA biomarkers detected by the assay include Single nucleotide variants (SNVs), insertions, deletions, multinucleotide variants (MNVs), gene amplification, gene deletion, gene rearrangements. TSO 500 also detects immunotherapy biomarkers for tumor mutational burden (TMB) and microsatellite instability (MSI). This clinical sequencing assay will be used for the patient enrolment in NCI clinical protocols and patient treatments.
Bioinformatics data analysis of TSO500 v1 sequencing has been using TSO500 local app v1.2 that was installed on the NIH Biowulf cluster. Now Illumina upgraded the assay panel with HRD and other new features into TSO500 v2. TSO500 v1 and analysis app v1.2 will be discontinued in later 2026. The TSO500 v2 requires TSO500 analysis software runs on Illumina Dragon Server. The analysis license is built in with TSO500 v2 reagent kit purchased.
With currently one Dargen Server is installed and running evaluation and testing on the NIH Biowulf cluster, we request to purchase and install the second Dragen Server to run analysis of clinical samples that are about 1000 cases per year.
3.0 OBJECTIVE
The main objective is to buy the bioinformatics data processing “DRAGEN” server (IT hardware) provided by the manufacturer (Illumina Inc.) for clinical Next Generation Sequencing. The DRAGEN server will have pre-installed Dragen TruSight Oncology 500 (TSO500) and Dragen TSO500 ctDNA analysis software support secondary analysis for the data generated by our current clinicalTSO500 assay, and new TSO500 v2, TSO500 HRD add-on assay, TSO500 ctDNA assay. The lab plans to start new TSO500 assays in July 2026. All new TSO500 assays come with Illumina’s proprietary bioinformatics pipeline that is only available on DRAGEN server. The server will be mounted on NIH Biowulf cluster by the NIH CIT team.
