Full Spectrum Flow Cytometer/Spectral Analyzer Sought Characteristics

A. Full spectrum flow cytometer (spectral analyzer)

a. In contrast to conventional flow cytometers in which peak emissions from excited fluorophores are detected in a limited number of individual channels, spectral analyzers use all detection channels for each fluorophore to produce a spectral emission signal. This provides improved flexibility in panel design, ability to re-use spectral references, and tools for removal of autofluorescence.

B. Multiple excitation lasers (at least five)

a. (355nm, 405nm, 488nm, 561nm, 637nm)

C. 147 detectors including 2 scatter parameters.

D. Spectral Reference Library

a. Ability to store single color reference data in a library which eliminates the need to run single color controls for different experiments.

E. Ability to mix and cool sample during acquisition.

F. Automated startup with performance QC.

G. Automated calibration and cleaning functions H. Ability to accommodate multiple plate/tube formats.

I. One year warranty.

Full spectrum flow cytometer (spectral analyzer) for characterizing human, animal, and bacterial cells using immunofluorescence methods. In contrast to conventional flow cytometry, the recent development of spectral flow cytometry offers certain advantages as it allows for simultaneous use of fluorophores of closely emitting spectra, incompatible with conventional flow cytometry;

expansion of immunophenotyping panel complexity to 40 parameters and beyond; and measurement of cellular autofluorescence as a separate parameter to avoid false-positive signals.

Primary work will entail characterizing major innate and adaptive immune cell populations from fresh blood using multicolor fluorescent panels which is currently beyond the capability of our conventional flow cytometer. We may also characterize cells from experiments using animal models, particularly mice. In addition, we will characterize immunoglobulin-coated bacterial cells from the human or animal intestinal microbiome to better characterize the status of the microbiome, and to characterize the immune response to these bacterial cells from the intestinal microbiome.
