The University of Washington will receive $1,876,269 under a three-year Project Grant award from the National Institutes of Health National Institute of Child Health and Human Development (NICHD) Child Health and Human Development Extramural Research program (CFDA 93.865). The grant will support research using Drosophila melanogaster to investigate motor control and premotor neural circuits in the spinal cord. Researchers will develop a behavioral task for flies to generate specific amounts of force to position the femur-tibia joint. The 12 motor neurons controlling the joint can be genetically labeled for targeted electrophysiological recordings to understand how the complete population functions dynamically. Electron microscopy will also be used to reconstruct central locomotor circuits and identify key premotor neurons. Recordings of these neurons during the task will reveal their contributions to processing sensory feedback and controlling leg force. Results aim to provide insights into how descending commands interact with internal models to control locomotion, with the long-term goal of informing interventions for neuromuscular disorders and engineered systems.
