Aim 1 integrates three-dimensional printing methods to enable spatial control and retrieval of cells and materials within the platform. Aim 2 establishes protocols for stable integration of a secondary endothelial cell monolayer atop the NIICHE and validates visualization of immune cell extravasation through the endothelium in response to chemokine gradients. Aim 3 expands the platform to additional T cell sources and evaluates differential immunogenicity of various cellular components. The NIICHE platform delivers three-dimensional, real-time, noninvasive measurements of immune cell recruitment and engagement with pancreatic beta cells, addressing limitations of current animal models and culture systems in understanding human T1D interactions and disease mechanisms. Work is performed in Gainesville, Florida. The period of performance runs through July 31, 2028.
