This Project Grant award from the National Institute of Neurological Disorders and Stroke (NINDS), under the Extramural Research Programs in the Neurosciences and Neurological Disorders (CFDA 93.853) program, will fund research at the University of California, Los Angeles (UCLA) to establish a zebrafish model for studying axon caliber regulation and determine if neurofilaments contribute to axon tapering. The $407,739 award, effective from August 1, 2025 to July 31, 2027, will support the following key objectives: 1) use live imaging to examine how microtubule dynamics, intracellular trafficking, and axon structure are affected by asymmetric tapering at branch points; 2) create fluorescent reporters to document the distribution of neurofilaments in peripheral axon arbors; and 3) characterize the effect of neurofilament deficiency on the tapering morphology of sensory neurons. This research aims to better understand the regulation of neurite caliber, which influences the electrical properties, trafficking capacity, and vulnerability of neuronal processes.
