Aim 1A tests whether podocyte-specific testican-2 deficiency increases kidney damage severity in murine models of focal segmental glomerulosclerosis and diabetic kidney disease, with assessment of kidney function markers (proteinuria, creatinine, GFR), histopathology, electron microscopy, and glomerular injury biomarkers. Aim 1A also uses podocyte-specific integrin-beta-3 null mice to determine whether kidney damage from testican-2 deficiency requires integrin-alpha-V-beta-3 activation. Aim 1B tests whether increasing circulating testican-2 levels can attenuate and rescue kidney damage in both testican-2-deficient mice and wild-type littermates using the same readouts as Aim 1A.
