The project uses fate-mapping and genetic mouse models to characterize how local immunological niche signals, particularly the CCL22-CCR4 axis in eosinophils, facilitate beige adipocyte generation and thermogenesis. Preliminary data indicate that CCL22 injection into adipose tissue promotes tissue beiging, while neutralizing antibody treatment blocks it, and that serum CCL22 levels correlate inversely with body weight and fat mass in both mice and humans. Performance takes place in Chicago, Illinois, with a period of performance extending from the award date through May 31, 2031. The project grant is a Research Grant assistance type supporting extramural basic biomedical research that improves understanding of disease mechanisms and leads to improved prevention, diagnosis, and treatment of metabolic and obesity-related disease.
