The project will characterize the role of microRNAs carried by hepatocyte-derived extracellular vesicles (HD-EVs) in controlling hepatocyte proliferation during liver regeneration. Using a rodent partial hepatectomy model, the research will identify the complete repertoire of inhibitory liver cell proliferation (ILCP)-active microRNAs in HD-EVs, map their messenger RNA targets and associated signaling pathways, and validate their mechanistic role in regulating liver regeneration. The work encompasses characterization of extracellular vesicle populations and their dynamic changes during regeneration, next-generation sequencing to profile microRNAs in HD-EVs and liver tissue, validation of ILCP activity in vitro and in vivo, and construction and experimental confirmation of microRNA-to-messenger RNA-to-pathway interaction networks. Performance will take place in Richmond, Virginia. The award period runs from September 1, 2026, through May 31, 2030.
