The research integrates imaging, bioinformatics, and cancer biology to identify novel molecular events across the genome, transcriptome, and epigenome in pediatric low-grade gliomas (PLGG), the most common central nervous system tumor among children. In Aim 1, the project leverages a clinicoradiomic risk model to classify newly acquired PLGG tumors from the Children's Brain Tumor Network into low, medium, and high-risk groups, then trains a preclinical multi-omic predictive model of drug response using data from the Childhood Cancer Model Atlas to identify potentially efficacious small molecules specific to each risk group. Multi-omic clustering will further subdivide risk groups to identify highly specific molecular aberrations to inform treatment. In Aim 2, the project functionally interrogates identified molecular targets using fruit fly and organoid models to test and validate effects of molecular events on PLGG progression and infiltration.
