Project Grant Award Summary The National Cancer Institute (NCI) awarded a $650,597 Project Grant to the University of California, San Diego, effective April 1, 2026, through March 31, 2031, under the Cancer Treatment Research program (CFDA 93.395). This research project investigates the role of B-cells in generating durable complete responses to checkpoint blockade immunotherapy in head and neck squamous cell carcinoma (HNSCC). The research aims to identify mechanisms underlying exceptional patient responses—approximately 3-5% of HNSCC patients achieve disease-free status for at least two years following immunotherapy treatment—and to develop strategies to enhance and refine immunotherapeutic approaches based on B-cell-mediated anti-tumor immunity. The project delivers research outputs organized across three specific aims: (1) determining the effects of checkpoint blockade immunotherapy on B-cell activation and testing novel agents that enhance B-cell-mediated anti-tumor immune responses; (2) identifying B-cell subtypes that mediate anti-tumor immunity and dissecting their mechanisms of action in orthotopic HNSCC models; and (3) defining the specificity and identifying novel B-cell antibody repertoires from patients with durable complete responses. The expected outcomes include enhanced understanding of B-cell involvement in complete immunotherapy responses and development of improved immunotherapeutic approaches to increase complete response rates in HNSCC treatment.
