Three specific aims guide the work: quantifying how interfragmentary strain regulates macrophage phenotypes and fracture fibrosis using a murine fracture model with tunable strain and longitudinal immune and histologic profiling; defining the role of macrophage-derived TSP1 in fibroblast activation under high strain using single-cell and spatial transcriptomics integrated with macrophage-specific THBS1 knockout mice; and rescuing regenerative healing by inhibiting TSP1 using the SRI-35241 peptide in macrophage-fibroblast co-culture and in vivo high-strain models.
