The project addresses mechanisms by which B. burgdorferi evades complement-mediated immune killing. The recipient has generated a CspZ mutant protein (CspZ-YA) selectively deficient in Factor H binding, which exposes previously hidden epitopes on the bacterial surface. Preliminary mouse studies demonstrate that CspZ-YA vaccination protects against Lyme disease infection, whereas wild-type CspZ does not. The project will characterize monoclonal antibodies recognizing the Factor H-binding sites on CspZ, establish mechanisms of complement-mediated protection, and develop CspZ-YA as a vaccine candidate targeting the most common Lyme disease causative agent in North America. Work is performed at North Grafton, Massachusetts. The period of performance runs from August 7, 2026, through July 31, 2031.
