This federal Project Grant award, funded by the National Institutes of Health (NIH) under the Allergy and Infectious Diseases Research program (CFDA 93.855), aims to develop a genomic surveillance approach for monitoring the emergence of artemisinin resistance in Plasmodium falciparum malaria parasites in Africa. The total funding award is $1,553,810.00 and the project period runs from December 1, 2024 to November 30, 2029. The key objectives of this research project are to: 1) identify parasite genetic loci under selection in four African endemic countries over time following the implementation of artemisinin-based combination therapies (ACTs); 2) validate the functional profiles associated with artemisinin resistance using genetic crosses and transcriptomic analyses; and 3) assess the population-level and within-host dynamics of selected alleles in Mali to inform future genomic surveillance strategies. The project will be conducted by the University of Maryland, Baltimore, with a sub-award to the University of Sciences, Techniques and Technologies of Bamako in Mali to support in-country research activities. The findings from this comprehensive genomic surveillance approach are expected to enable earlier detection of emerging artemisinin resistance in Africa, before it manifests clinically, and provide critical insights to guide the design of future resistance monitoring efforts.
