The grant supports three specific research aims: (1) developing and optimizing novel nanoparticle formulations to inhibit alcohol-related macrophage activation and hepatic stellate cell transformation through targeting of carcinoembryonic antigen (CEA) signaling, microRNA-155, and the chemokine receptor CXCR4; (2) characterizing the effectiveness of the anti-CRLM nanoparticles and assessing the contribution of metastasis-promoting macrophages through ex vivo and in vivo assessments; and (3) performing transcriptomic analysis to determine the intrahepatic macrophage gene signature, localization, and prometastatic properties.
