- The National Cancer Institute awarded ST Jude Children's Research Hospital Inc. $737,023 under the Cancer Treatment Research program (CFDA 93.395) on July 23, 2026, to develop and evaluate CD19-CAR T cell therapy combined with TCR alpha/beta–depleted haploidentical hematopoietic cell transplantation for pediatric patients with relapsed or refractory CD19+ acute lymphoblastic leukemia and lymphoma. The research aims to minimize relapse risk and graft-versus-host disease while eliminating total...
- The National Cancer Institute awarded The Children's Hospital of Philadelphia Research Institute $276,545 on June 8, 2026, under the Cancer Research Manpower program (CFDA 93.398) to develop genomic risk prediction models for T-cell malignancies. The recipient will identify high-risk genomic features associated with relapse and death in T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoblastic lymphoma (T-LBL) stratified by genetic ancestry, then develop and validate risk prediction...
- The National Cancer Institute awarded The Regents of the University of California, San Francisco $172,889 on January 12, 2026, under the Cancer Research Manpower program (CFDA 93.398) to develop and optimize chimeric antigen receptor (CAR) T-cell therapies for acute myeloid leukemia (AML). The funded research applies structure-guided protein evolution to improve natural ligand-based CAR-T therapies targeting CD70 in AML. The project uses computational deep learning algorithms and high-throughput...
- The National Cancer Institute awarded The Children's Hospital of Philadelphia $738,666 on August 4, 2026, under the Cancer Treatment Research program (CFDA 93.395) to engineer CD19 mimotopes for enhancing and repurposing CD19-CAR T cell therapy against hematological malignancies and solid tumors. The recipient will develop a high-affinity CD19 mimotope identified through yeast surface display screening to pair with existing CD19-CAR T products. The linear mimotope, a 10 amino acid peptide, is...
- The National Cancer Institute awarded The Johns Hopkins University $218,089 on August 1, 2026, under the Cancer Treatment Research program (CFDA 93.395) to develop and test engineered natural killer cell therapy targeting acute myeloid leukemia. The funded work develops chimeric cytokine receptor (CCR)-engineered NK cells designed to attack AML-associated surface antigens and overcome AML-mediated NK cell suppression. In Aim 1, the recipient will engineer NK cells to express dual interleukin...
- The National Cancer Institute awarded The Trustees of the University of Pennsylvania $434,969 on May 1, 2026, under the Cancer Treatment Research program (CFDA 93.395) to investigate type I interferon response and differentiation in FLT3-inhibitor persistent acute myeloid leukemia. The funded research develops genetically engineered mouse models of acute myeloid leukemia with inducible and reversible FLT3 mutations to understand cellular mechanisms that maintain FLT3-mutant clone persistence...
The National Cancer Institute awarded The Children's Hospital of Philadelphia $263,656 on August 17, 2026, under the Cancer Research Manpower program (CFDA 93.398) to support research into novel mechanisms of acute myeloid leukemia (AML) resistance to chimeric antigen receptor T cell (CAR-T) therapy. The research aims to develop a detailed understanding of how altered STAT5 levels dictate poor outcomes for AML CAR-T therapies and to identify a combination therapy that selectively blocks AML resistance signaling via CD131 while preserving CAR-T anti-leukemia effect. Recent clinical trials of CAR-T targeting CD123 on AML blasts revealed an unexpected mechanism of AML resistance whereby activated CAR-T cells secreted cytokines (GM-CSF and IL-3) that support AML survival through CD131 receptor signaling via JAK/STAT pathways. Blocking this cytokine-mediated resistance with broad JAK pathway inhibitors may impair CAR-T cells in humans; the research will identify clinically actionable approaches to safely overcome this AML survival advantage and develop effective CAR-T therapies for AML. The award supports investigator-initiated research at The Children's Hospital of Philadelphia Research Institute in Philadelphia, Pennsylvania, with a period of performance through July 31, 2031. The Cancer Research Manpower program provides training and research support to nonprofit and for-profit institutions offering biomedical training opportunities for individuals pursuing careers in basic, clinical, and prevention research important to the National Cancer Program.
