Description

Abstract
Microbial development of antibiotic resistance in recent decades poses a serious threat to worldwide health and necessitates new approaches towards antibiotic discovery. In response, recent research initiatives have turned to discover new leads from microbial genomes, which harbor vast libraries of genes encoding untapped metabolic diversity. Betalactones are a small subclass of natural products which display potent antimicrobial activity. However, little is known about the biosynthetic pathways responsible for their production. Here, we propose a new approach towards discovery of active betalactones based on functional genomics. We plan to mine microbial genomes obtained from a marine sponge for betalactone-producing gene clusters. Once selected, we will characterize the enzymes involved in betalactone biosynthesis and their substrates. Our two-pronged approach elucidates both bioactive molecules and the enzymes that produce them, thus providing for scalable metabolite production in heterologous strains. In addition, this proposal evaluates the potential of genomically-discovered compounds to function as antibiotic leads. Importantly, this genomics-based strategy allows for the discovery of natural products with minimal prior knowledge on their structure and the species that produce them. The research outlined in this study is planned in a three-year program which will take place in the laboratory of professor michael d. Burkart at UC SAN diego (described under training activities and timeline). Our laboratory is well-equipped with the biochemical, chemical, and molecular biological tools and expertise necessary for completion of the proposed aims.
