Description
- Abstract
- Changes in 3' end processing of RNA are implicated in many diseases, including several malignancies. A highly conserved hexameric polyadenylation signal sequence (PAS) directs processing machinery where to cleave nascent RNA to add a polya tail and is central to the mechanism by which 3' ends are processed. The transcript coding for antibodies contains two terminal exons that code for a transmembrane domain, which is flanked by two PAS, one encoding membrane(MIG), and the other encoding secreted (SIG). We hypothesizeis regulated by trans-acting factors that either interact directly withrna or change the transcriptional landscape of the cells. Using RAP-MS2.0, a method developed in our lab we have identified 80 proteins conserved between mouse and human b cells that interact directly withrna. We intend to validate these interactions using biochemical methods to map the interactions and functional genomics (aim i). To understand how the transcriptional landscape contributes to APA in b cells, we will use a reporter-based, high throughput screen of TFS to identify regulators of the MIG to SIG transition. (aim II). Through these complementary aims, we expect to identify the factors that mediate this mechanism at the crux of autoimmunity and cancer. Furthermore, the central product of this grant is my interdisciplinary training as a graduate student, which involves multi-layered mentoring to promote my growth towards an academic career with categorical focus on disciplinary, professional, and pedagogical development.
