The National Science Foundation awarded $564,893 under the Mathematical and Physical Sciences (CFDA 47.049) federal grant program to the University of Wisconsin-Madison to develop a chemically defined, plant-derived biomaterial platform for human cell culture from September 1, 2022 to August 31, 2025. The award will support research to decellularize plant leaf material for use as a broadly useful and adaptable biomaterial for human cell culture. Researchers will characterize the chemical composition, cytotoxicity, morphology, degradability, mass transport and mechanical properties of the leaf material. Conformal polymer coatings will be developed and optimized to enable human cell adhesion, viability, alignment and stem cell differentiation toward tenocytes and myocytes. The effect of leaf microtopography and customizable surface chemistry on cellular alignment and differentiation will be systematically evaluated. Outcomes will include a decellularization process and comprehensive biomaterial characterization, as well as novel copolymer coating chemistry to functionalize the plant-derived scaffold for human cell applications.
