This $225,000 Project Grant from the National Science Foundation Division of Molecular and Cellular Biosciences will fund research into the detailed characterization of topologically associating domain (TAD) boundaries in eukaryotic genomes. The grant supports the Biological Sciences program, which aims to increase scientific knowledge of major biological problems. The Trustees of the University of Pennsylvania, doing business as the Clinical Practices of the University of Pennsylvania, will conduct novel single-molecule genomics approaches and super-resolution imaging to identify and quantify the genetic elements that insulate neighboring TADs in single cells. This could help update models of TAD formation and improve understanding of how boundaries contribute to TAD structure, dynamics and function. Findings may also explain how TAD structure can be reorganized by changes to the non-coding genome. The research will provide cross-disciplinary training for graduate and undergraduate students. Results have the potential to advance knowledge of gene regulation, DNA replication, repair and recombination mechanisms.
