This $650,000 federal Project Grant award from the USDA's Agriculture and Food Research Initiative (CFDA 10.310) will support research by Utah State University (USU) to investigate alternative mechanisms of pregnancy recognition in ruminants beyond the established role of interferon-tau (IFNT) signaling. The key products and services to be delivered through this 4-year project include: Characterizing differences in gene expression within the trophoblast, endometrial epithelial cells, and corpus luteum of wild-type versus biallelic IFNAR2 knockout sheep that can maintain pregnancy without IFNT signaling. This analysis will be conducted using single-cell RNA sequencing. Identifying potential antiluteolytic or luteotropic agents present in the uterine luminal fluid and secreted by cultured trophoblast cells, using liquid chromatography-mass spectrometry. The findings from this research are expected to revise the accepted paradigm for pregnancy recognition in ruminants, as the project explores an alternative mechanism that allows for normal pregnancy in the absence of IFNT signaling through the type I interferon receptor.
